Objective:Present work aims to observe the expressions of Desmin and dystrophin in levator ani muscle with pelvic floor dysfunction (PFD), to detect biochemical changes in levator ani muscle, and to explore whether the alteration in Desmin and dystrophin in levator ani muscle tissue expression could contribute to pelvic floor dysfunction;Methods:Biopsy specimens of levator ani muscle were obtained from30patients with SUI,30patients with POP and6asymptomatic control with rectal cancer during operation. Levator ani muscle’s structure was examined with routine histological techniques. Real-Time PCR was used to quantitatively determining the mRNA level of Desmin and dystrophin mRNA;Results:1. Real-time PCR(1) Compared with the control group, the expressions of dystrophin and Desmin mRNA in group POP and group SUI were lower, this difference is of significance to statistics (p<0.001).(2) The expressions of Desmin and Dystrophin in levator ani muscle mRNA in the group POP and group SUI has no statistical significance (P>0.05).2. Western Blot detect(1) Compared with the control group, the expressions of dystrophin and Desmin mRNA in these two groups had reduced, the difference is statistically significant (p<0.001).(2) Compared group POP with group SUI, there is no significance to statistics in terms of the expressions of Dystrophin in levator ani muscle,(p=0.102).(3) Compared the Desmin in the two groups, group SUI is lower than group POP, this difference is significance to statistics (p<0.001).3. The severity of disease, the parity have the linear regression relationship with the expressions of mRNA in Desmin and dystrophin. Ages and menopause have no relations with the expressions of Desmin and dystrophin mRNA.Conclusions:1. The occurrence of female pelvic floor functional disorder is related with the reduction of the expression of dystrophin and Desmin in female levator ani muscle.2. The severity of disease has a linear regression relationship with the expressions ofDesmin and mRNA in dystrophin, and is negatively correlated. The worse of the pelvic organ prolapse and stress urinary incontinence, the less of contents of Desmin and Dystrophin in the tissue of pelvic floor muscles.3. The parity has a linear regression relationship with the expressions of Desmin and mRNA in dystrophin, and is negative correlation. That means the more times of delivery, the expressions of Desmin and dystrophin in levator ani muscle are lower.
