| objective:To investigate the effect of periurethral injection of exogenous inechano-growth factor(MGF) on the expression of endogenous MGF mRNA during repair and regeneation process of the injured urethral muscle in female wistar rat.Methods:90female virgin wistar rats were randomly divided into3groups:such as blank group (without injury and without injection)、control group (injection with physiological saline)、experiment group (injection with MGF-Ct24E),30/group.Control group and experiment group were made model of urethral muscle injury by intravaginal balloon inflation,then,the rats of experiment group were taken periurethral injection of rat MGF-Ct24E in the middle urethral muscle and the control group rats underwent periurethral injection of physiological saline,the blank group rats is without intravaginal balloon inflation and injection. The samples(n=6/group) were taken on the1、4、7、10、14day respectively and the middle urethra muscle were processed for SYBR Green RT-PCR. The statistical analysis adopt factorial analysis of variance design data. P<0.05is considered that difference is statistically significant.Results:The expression of MGF mRNA in the blank group had no significant difference in statistics(P>0.05)at the each time.At the each time following injury,as compared with the blank group,the expression of MGF mRNA in the control group increased significantly at day4、7、10(P<0.05); The expression of MGF mRNA in the experiment were significantly higher than the control group at day1、4、7、10,(P<0.05); The expression of MGF mRNA in the control group reached the peak on day7,but the experiment group peaked at day4,and then,the expression of MGF mRNA begun to drop gradually between two groups;The experiment group’s MGF mRNA level still higher than that of control group at day14,but had no significant difference in statistics(P>0.05).Conclusion:The expression of endogenous MGF mRNA was facilitated and maked its peak in advance by periurethral injection of MGF Ct-24E during repair and regeneration period following urethral muscle injury in female rat. The findings suggest that the repair of impaired urethral muscle may be improved by periurethral injection of MGF Ct-24E. |
