| Objective: Through observating the effects of Kangaifangzhuan Decoction onmouse hepatoma lymphatic metastasis, and detecting expression of FAK, p-FAK andPI3K, p-PI3K,study the mechanism that the decocion inhibits murine hepatocellularcarcinoma lymphatic metastasis.Methods:615mice were randomly divided into control group, positive controlgroup, treatment group1(low dose) and treatment group2(high dose),in order toestablish the mouse hepatoma lymphatic metastasis model with inoculation method.Regular observating the body weight changes of mouse.After21days,the mouse withinoculation was killed,and detecting the weight of mouse solid tumor and lymph nodemetastasis.Applicating the immunohistochemistry to detect the expression level ofFAKã€p-FAKã€PI3K and p-PI3K of the primary tummor of each group.Using SPSS11.0statistics software to analyse the datas.Result:In tretment groups, compared with the postitive control group,the weightof mouse had been gained,the tumor weight and the incidence of lymph node metastasisin mice had been declined(p<0.05).The expression level of FAKã€p-FAK and p-PI3K inprimary tumors are reduced(p<0.01). Westernblot detection results of metastases tuomrin FAK, p-FAK and PI3K, p-PI3K is not well.Conclution:Kangaifangzhuan decoction suppress the lymphatic metastasis ofhepatocarcinoma in mice, it is relation with the FAK/PI3K signaling pathway... |
