Objective: To investigate the clinical significance of expression of Sonic-Hedgeho(gShh)and Stathmin in human benign prostatic hyperplasia,low grade prostaticintraepithelial neoplasia(LGPIN) and prostatic carcinoma tissues.Methods: The expression of Shh protein and Stathmin protein are detected in30cases of human benign prostatic hyperplasia tissues,12cases of low grade prostaticintraepithelial neoplasia(LGPIN) and30cases of prostatic carcinoma tissues withimmunohistochemistry method.We also detected the expression of Shh mRNA andStathmin mRNA in15cases of human benign prostatic hyperplasia tissues and15casesof prostatic carcinoma tissues with RT-PCR method.Results:1.Immunohistochemistry:The positive expression rate of Shh protein were36.67%(11/30),60%(6/10) and73.33%(22/30) respectively detected in human benignprostatic hyperplasia,LGPIN and prostatic carcinoma tissues, the positive expressionrate of Stathmin protein were23.33%(7/30),41.67%(5/12)and83.33%(25/30)respectively detected.The expression of Shh and Stathmin were similar in human benignprostatic hyperplasia tissues and LGPIN tissues(P>0.05),but the expression of Shh andStathmin were higher in the group of prostatic carcinoma than that of in othergroup,there was a significant difference between three groups(P<0.01).2.RT-PCR: The expression of Shh mRNA and Stathmin mRNA were higher in thegroup of prostatic carcinoma than of in benign prostatic hyperplasia,the expression ofShh mRNA and Stathmin mRNA in prostatic carcinoma were markedly highercompared with benign prostatic hyperplasia tissues at mRNA level(P<0.05).3.The expression of Shh protein or Shh mRNA has no correlation with PSA level and preoperative prostate weight calculated by B-ultrosonography(P>0.05), There is asingnificant difference between clinical stage and gleason score(P<0.05). As is the casewith Stathmin.4.The data shows that maybe there is a relation between Shh and Stathmin(P<0.05).With the elevated level of Shh,there is a significant increase in the expressionin the Stathmin.Conclusions:1.The Shh and statmin were not expressed or expressed lower in human benignprostatic hyperplasia tissues and LGPIN tissues,but higher in prostatic carcinomatissues.2.The expression of Shh had no correlation with the PSA level and prostateweight,but there is a singnificant difference within clinical stage and gleason score,thepositive rate of Shh or Stathmin increased with the aggravation malignant and the highstage.3.The Shh and statmin were expressed lower in LGPIN tissues,it parallels that inbenign prostatic hyperplasia tissues.In these patients,the incidence of prostatic cancer islower.4.The expression pattern of Stathmin parallels that of Shh and suggests that maybethere is a relation between Shh and Stathmin,the interactions may play an important rolein prostatic disease progression.
