Coronary heart disease is a serious threat to human health, common diseases andfrequently-occurring disease, atherosclerosis, their lesions base from the currentresearch atherosclerosis is a chronic inflammatory process involved in a variety offactors, so the lesionfound to have varying degrees of inflammatory cell infiltration, andmost important macrophage infiltration。The pit is the invagination of the plasmamembrane of the cell membrane of a cell surface specific micro area, mainly composedof cholesterol, glycosylated sphingolipids, sphingomyelin, and caveolin is an importanthub of cell signaling in the cell membrane region。The study found that small concaveand cav-1can be expressed in inflammatory cells, damage and chronic inflammationsuch as atherosclerosis, cav-1expression in the normal coronary high expression instable plaque macrophagessmall, unstable plaque macrophages express at least, canregulate TNF-а, MMP-1expression in coronary plaque macrophages.Objective: In this study, by immunohistochemical methods to detect cav-1, TNF-а,MMP-1in human coronary sclerosis stable plaque and unstable plaque groupmacrophages express observed cav-1expression and TNF-а, MMP-1expression andatherosclerosis progress in the degree relationship.For the study of human coronary atherosclerosis pathogenesis and targets for thetreatment of experimental evidence.Methods:60coronary artery specimens obtained from autopsy specimensaccording to the stenosis degree on the light microscope after HE staining, inaccordance with whether the lipid core and its size is more than40%of the area whereplaque, the specimens were divided intostable plaque group (24cases), unstable plaquegroup (21cases), and normal control group (15cases). Method usingimmunohistochemical detection of cav-1, TNF-а, MMP-1expression in coronary plaque macrophages situation. And marked with CD68macrophages。Results SPSS13.0software package for statistical analysis, monitoring indicators Wilcoxon test was usedand pairwise comparisons using Pearson correlation analysis of the relationshipsbetween them.Result:1.cav-1as the main component of the pit structure on the cell membraneexpression of the positive parts in the cell membrane and cytoplasm of normal coronaryendothelial cells, smooth muscle cytoplasm and cell membrane. Less stable plaquegroup of macrophage infiltration cav-1high expression and unstable plaque group ofmacrophage infiltration cav-1low expression or negative expression, the expression ofdifference was statistically significant between the stable group and the unstable group.2.TNF-а, expressed in the normal coronary little or negative expression, part of thestable plaque macrophage expression of highly expressed in macrophages in unstableplaques, positive parts of the cytoplasm.3.MMP-1expression in the normal coronary group with little or negative,macrophages in stable plaque group, part of the expression of highly expressed inmacrophages in the unstable plaque group, positive parts of the cytoplasm.4.Non-parametric rank correlation test statistical analysis to show cav-1, TNF-а,the expression intensity of MMP-1in atherosclerotic plaques was negatively correlated.Counciusion:1、No infiltration of macrophages in the normal coronary group, lessmacrophage infiltration in stable plaque group, infiltration higher in the unstable plaquegroup2. cav-1expression in macrophages in stable plaque group was significantlyhigher than the unstable group, TNF-а, MMP-1expression in macrophages in stableplaque group lower than the unstable group. The Cav-1, TNF-а, MMP-1expressionsuggests that they are involved in the coronary atherosclerotic lesion formation anddevelopment of one important factor, but also play an important role in promotingplaque instability.3、cav-1low expression of coronary atherosclerosis can promote thedevelopment of atherosclerosis lesions, by promoting the infiltration of inflammatorycells and factors so that the increase in plaque instability, leading to plaque prone torupture, bleeding. However, because this experiment only from unilateral explore, andtherefore more limited, more in-depth and detailed discussion from multipleperspectives.
