Backgrounds: Recently genetic association study and genome-wide associationstudy identified one SNP rs1739843in HSPB7gene that showed significant associationwith idiopathic dilated cardiomyopathy (IDC) in multiple populations of Europeanancestry.But it is unclear whether rs1739843is associated with idiopathic dilatedcardiomyopathy in Chinese Han population.Objective: The aim of the study was to test whether rs1739843is associated withIDC in a different ethnic population, namely a Chinese cohort.Methods: The study was a case-control study which included306IDC patientsand872control subjects. All the subjects were Chinese Han population and came fromDaLian Medical University affiliated first hospital and Wuhan Xiehehospital,2008-2011. The criteria of IDC patients were based on the standard diagnosticcriteria according to AHA2006guidelines. The criteria were:⑴theleft ventricularend-diastolic diameter (LVEDd)>50mm(female) or>55mm(male);⑵the left ventricularejection fraction (LVEF)<45%and/or the left ventricular fractionshortening(LVFS)<25%.⑶t he obvious causes which could lead to IDC such as hypertension,coronaryartery diseases,,valvular disease, congenital defect, alcoholic cardiomyopathy,tachycardia-induced cardiomyopathy or peripartum cardiomyopathy et al wereexcluded;⑷p atients with a positive family history were also excluded.Age,sex andbasic diseases(such as hypertension and diabetes) of the control group were matchedwith the case group and excluded the IDC patients. We collected blood samples of twogroups of subjects to extract DNA(Beijing TianGen biological corporation)andsearched the gene sequence of rs1739843from SNP database of Pubmed database. Then we designed primers of rs1739843and amplified the desired gene fragment. rs1739843was genotyped using fluorescent dye-based high-resolution melt analysis on aRotor-gene6200System (Corbett Life Science)according to the protocols from themanufacturers.Results: rs1739843showed significant association with IDC in a Chinese Hanpopulation(p=0.022).After corrected age, gender, diabetes mellitus and hypertensionby logistic regression analysis,the association still maintained significant(p-adj=0.020,OR=0.782),and minor allele T was a protect allele;in a dominant model,rs1739843andIDC showed significant association(p=0.046);after corrected age, gender, diabetesmellitus and hypertension presented association (p-adj=0.041,OR=0.778);in a additionmodel, it presented no association between the two aspects(p-adj=0.07), but aftercorrected rs1739843was associated with IDC ((p-adj=0.024,OR=1.327).Conclusions: HSPB7gene single nucleotide polymorphism rs1739843wasassociated with IDC and minor allele T was a protect allele.
