Objective:To explore the mechanism of curcumin in tumor tissue. Which can provide new ideas and new experimental evidences for clinical treatment of androgen independent prostate cancer.Methods:1、To develop a nude mice subcutaneous transplantation model of human prostate carcinoma with cell Line PC-3. According to the volume of transplantation tumor.2、The12nude mice were randomly divided into two grounds, there was6nude mice in each group. Curcumin group:daily intraperitoneal injected(100mg/kg), a total of4weeks.. At the same time, Negative control group:intraperitoneal injection volume containing6%(by volume) aqueous solution of polyethylene glycol containing6%(volume fraction) ethanol.3、To observe the situation of nude mice every day, and produce the tumor volume growth curve by measure tumor volume. Calculate inhibition rate of the tumor through to get the tumor after the experiment.4、HE stained to observe the morphological characteristics of tumor. Immunohistochmistry and western blotting analysis was used to examine the expression of FASN, Shh and Bax proteins in the tumor tissues of nude mice. Explore the mechanism of curcumin in tumor tissue. Results:1. Model was successfully established in nude mice with human prostate cancer PC-3cell.2. In nude mice experiments, the tumor volume and weight in curcumin group significantly reduce (P<0.05) compared to the negative control, and the tumor growth was suppressed obviously with52%of on costatic rate.3. In tumor tissue, HE stained tumor cells apoptosis and immunohistochemical examination revealed that the protein expression of FASN and Shh was decreased, but Bax was increased. Western blotting quantitative detection the expression of FASN and Shh in curcumin group significantly reduce (P<0.05) compare to the negative group. The expression of Bax in curcumin group was significantly rise (P<0.05) compare to the negative group.Conclusions:1. Curcumin could significantly inhibited the transplanted tumor growth in nude mice model of PC-3cell, reducing the tumor volume and weight of tumor.2. Curcumin remarkably restrained the expression of FASN and Shh protein in tumor tissues and increased the expression of Bax protein in the tumor tissues.3. The mechanism of Curcumin inhibit transplanted tumor growth of prostate cancer PC-3cell in nude mice may be related to downgrade the expression of FASN and Shh protein and increase the expression of Bax protein.
