Objective To investigate the effects of Chenxiang Huaqi capsule on the intestinalICCs and myenteric plexus in diabetic rats.Methods The healthy male SD rats were divided randomly into three groups: Controlgroup, diabetes mellitus(DM)group and DM+Chinese Herb group (DM+CH group).Diabetic rats models were established by intraperitoneal injection of streptozotocin(STZ,60mg/kg, ip) one time, then DM+CH group were given herb15.75g/(kg·d), DMgroup and Control were given an equal volume of distilled water at the same timeeveryday. All rats after4weeks intervention, were given Indian ink by lavage for thesmall intestinal transit rate determination. The number and immune response intensityof c-Kit, PGP9.5, NFP and CBS-positive products were studied byimmunohistochemical method and quantified image analyzer.Results After four weeks, small intestine transit rate of DM+CH group was increasedobviously (71.26±5.22vs45.52±6.42, P <0.01) compared with DM group, still reducedobviously compared with Control group (71.26±5.22vs80.40±7.33, P <0.05); the totalarea and the mean optical density value of c-Kit-positive products were significantlyincreased in the myenteric plexus of DM+CH group compared with DM group(443.28±24.40vs358.83±35.03, P <0.01;0.16±0.02vs0.13±0.02, P <0.01), but stillreduced compared with control (443.28±24.40vs557.28±42.35, P <0.01;0.16±0.02vs 0.18±0.02, P <0.05), and so did PGP9.5and NFP. Whereas the total area and the meanoptical density value of CBS-positive products were significantly reduced in themyenteric plexus of DM+CH group compared with DM group (417.03±53.76vs484.20±77.80, P <0.01;0.20±0.03vs0.27±0.03, P <0.01), increased compared withcontrol (417.03±53.76vs340.57±78.23, P <0.01;0.20±0.03vs0.14±0.03, P <0.05).Conclution Chenxiang Huaqi capsule may improve the intestinal motility in diabeticrats by inversing partially abnormal changes of the intestinal ICC, PGP9.5, NFP andgaseous transmitter H2S. Using immunohistochemistry, reversetranscription polymerase chain reaction (RT-PCR) and western blot analysis,synaptophysin(Syn) expression were measured in myenteric plexus in Control group,DM group and DM+CH group. β-actin,a steadily expressed gene, was used as theinternal reference. The ratio of Syn mRNA、Syn protein to β-actin mRNA、β-actinprotein in the integrated optical density was calculated to represent the relativeexpression amount of Syn mRNA and Syn protein in the tissues.Resutls After four weeks, small intestine transit rate of DM+CH group was increasedobviously (71.26±5.22vs45.52±6.42, P <0.01) compared with DM group, still reducedobviously compared with Control group (71.26±5.22vs80.40±7.33, P <0.05); the totalarea and the mean optical density value of syn-positive products were significantlyincreased in the myenteric plexus of DM+CH group compared with DM group (832.33±58.78vs488.83±58.56,0.25±0.02vs0.16±0.01, P<0.01), but still reducedcompared with control (832.33±58.78vs937.67±101.23, P<0.05,0.25±0.02vs0.29±0.03, P<0.01), the expressions of Syn mRNA and protein were increased inDM+CH group as compared with that of DM group (0.107±0.010vs0.085±0.011,P<0.05,0.418±0.026vs0.330±0.023, P<0.01), but reduced compared by controlgroup(0.107±0.010vs0.255±0.011, P<0.05,0.418±0.026vs0.518±0.044,P<0.01).Conclutions Chenxiang Huaqi capsule may improve the intestinal motility in diabeticrats by inversing partially abnormal changes of the intestinal synaptophysin.
