[Objective] To investigate the expression of Derlin-1in the lung tissues in therat model of cigarette smoke-induced COPD.[Methods] The entire experiment was divided into two parts: animal researchand cell research.1Animal researchForty adult male Wistar rats were randomly divided into four groups:2-month CS exposed group,4-month CS exposed group,smoking cessationgroup(exposed to4months CS and then smoking cession for1month),control group.We adopt a passive exposing with CS to established the model of COPD. Collectingthe control and the experimental groups’ lung tissues,use Reversetranscriptase-polymerase chain reaction(RT-PCR) to detect the mRNA expression ofDerlin-1and use immunohistochemistry and Western blot to detect the proteinexpression of Derlin-1.2Cell researchPrimary and subcultured of rat AECII; CSE preparation; at0h,3h,6h,12h,24hand48h time point of rat AECII exposed to10%CSE: RT-PCR and Western blot wereused to detect protein expression levels of Derlin-1.[Results]:1.The result of animal research(1) In the control rats,the lung sections maintained the normal pulmonarystructure,while in the2-month CS exposed rats,the lung structure appears disorder andsome alveolar rupture.In the4-month CS exposed rats,lung structure was badlydamaged,with ruptured or thinned alveolar walls.The damaged of the alveolar structure was also obvious in the smoking cessation group.(2) Immunohistochemical results showed that Derlin-1is mainly located incytoplasm of vascular endothelial cells,alveolar epithelial cells and bronchialepithelial cell.The expression of Derlin-1is the highest in rats after exposure to2months CS,followed by the ex-smoking rats,and the lowest in controlgroup.Comparing to the smoking cessation group,the expression of Derlin-1isdecreased in rats exposed to4months CS.The result of RT-PCR and Western blottingis consistent with immunohistochemistry results.2The result of cell research(1) Western blot results revealed that in AECII exposed to10%CSE,Derlin-1protein expression is increased gradually at first, then gradually declined. Derlin-1protein expression is peak in6h group,then declined by degrees but still higher thancontrol group.The results of RT-PCR were consistent with Western blot results.[Conclusion]:1.With the prolonged of the cigarette smoke exposure time, the expression ofDerlin-1in lung tissue appeared corresponding changes.2. Derlin-1may be involved in the occurrence and development of chronicobstructive pulmonary disease。...
