Objective:This work was undertaken to determine the effect of neurotrophin-3when wastogether with neural stem cell in promoting neural stem cells into the proliferation anddifferenttiation of orientation, to detect if it can be better than a simple NSC transplantation inHIBD nerve functional recovery.Methods: Brain tissue was get from the wistar rats womb or newborn rats, which wascultured and then identified to make sure that were neural stem cells,and Made from singlecell suspension, waiting for transplantation. A total of78Wistar rats were divided into fourgroups: normal group, HIBD group, the NSC transplantation group and the NT-3combinedtransplantation group. Used the left common carotid artery ligation+anaerobic2hours intoHIBD model in7days after birthed, normal group which have no interruption.3days afterthe induction of HI, the three groups were given PBS, neural stem cells, NT-3joint NSCs.Continue to feed after transplantation, At the3th and7th week after surgery, The balancebeam test were performed respectively. And at last, all the rat brain were obtained Hema-toxylin and eosin staining and immunohistochemistry stain was performed latter. At the sametime, Ultrathin sections were performed for four group.Results: Suspended growth round ball or irregular round ball cell mass in vitro culture fetalbrain can be seen, Nestin-postive suggests it is neural stem cells; Behavior observation: HIBDrat express backwardness indefinite, explore behavior and double side hind leg joint activitiesstiffness on the balance beam; And neural stem cells transplantation group is activity obviousmore recovery than HIBD group joint, NT-3groups were more apparent. Statistical datashows that4weeks NT-3groups and NSC group comparison (P <0.05) have statisticalsignificance. Pathology observation: HE: Normal tissue of ventricle side was damaged andmore irregular cystic cavityin HIBD group,But In NSC transplantation group and the NT-3combined transplantation group,Normal tissue of ventricle side structure resumed, capsulebecome rare. Immunohistochemistry: MBP positive cells of NT-3joint group visible wassignificantly increased, and structure of cell array obviously improved. At high magnificationobservation MBP positive cell number group were14.83±2.44a、3.13±0.39a、7.04±0.93a、12.87±1.19a,the statistical analysis results: NT-3groups and NSC group comparison (P<0.05) have statistical significance. And GFAP staining NT-3group positive neurocytes wassignificantly reduced, and NSC group of comparative statistics still have obvious difference.Under the electron microscope, from cell ultrastructure level observed Ultrastructure hasimproved significantly and the myelin sheath fiber array obviously improved, organellesarrangement slightly owe rules, Relatively simple NSC transplanted group also has a distinctimprovement.Conclusion:NT-3can promot the proliferation of neural stem cells and induce neural stemcells to oligodendrocytes, which can promote the functional of rats ischemic hypoxic brainrecovery effectively.
