Chronic Prostatitis(CP) is a common disease in adult men, The main clinical manifestations is chronic pelvic pain, the mechanism of the disease is still unknown,Which makes its long-term effect of relieving pelvic pain is not good.At present,the mechanism of the disease has many theories.however,any theory can’t explain the pathogenesis and development of prostatitis completely.nowadays,the mechanism focuses on multiply theory,including infectious factors, immunological factors, endocrine factors and Mental psychological factors and so on,these factors interact with each other and result in the disease. With the development of molecular biology and immunology,The immune theory become the focus of basic research.With unique efficacy in the treatment of chronic prostatitis using Chinese medicine, Chinese medicine has a certain advantage.YouFang Liu who is a famous expert in combination of Chinese and Western medicine in urology in our country invented QianLieXian decoction (Renamed Danpu capsules)in the last century60s based on main symptoms local pain such as hypogastric zone.the waist and perineal position and prostate hardening,tenderness by digital rectum examination(DRE),it is line with syndrome of blood stasis of TCM and on pathological change feature of prostate-obstruction of glandular cavity, stagnation of prostatic fluid and interstitial fibrosis through combining macroscopic syndrome and microcosmic syndrome differentiation.My tutor,Prof YaQinag Zhang.is the academic heir of Prof YouFnag Liu.He makes a penetrating study to explore molecular and immune mechanism through clinical and experimental research on the basis of taking charge of scientific research topic from NSFC,state administration of TCM. clinic trial of Danpu Capsule shows that it can be obviously improved the quality of life by relieve pelvic pain in patients. We found that it can regulate the immune function through animal experiments, However the mechanism of Danpu Capsule relieving chronic pelvic pain is still unknown,this experiment is resarching cytokines related with pain through improved EAP rat model and revealing the mechanism of Danpu Capsule treating prostatitis.Purpose:The study aims to establish improved EAP to explain the immunologic mechanism of DP capsule treating CPPS through affecting the pain fators such as NGF,IL-6,β-EP in improved EAP rat model.Method:Adopting pure rat prostate protein and complete Freund’s adjuvant produce homogenate,then using the homogenate injects the prostate combining Incomplete Freund’s adjuvant injecting under the skin is to make rat model of EAP.Wistar rat were divided by6groups:high dose group of DP capsule(HD),middle dose group of DP capsule(MD),low dosage group of DP capsule(LD),Qina Lie Tai pill group(QLT),the model group and the normal group. They were sacrificed8weeks later,the gross shape was viewed by microscopic observation.Finally,the level of IL-6,NGF,β-EP in serum and prostatic tissue were detected in different groups by means of ELISA and RT-PCR..Result:1.ELISA testing NGF express level:the level of NGF in the prostate tissue and serum of the model group is on the rose, compared with the normal group,differences are statistically significant (P<0.01).after medical treatment, the level of HD,QLT in the prostate tissue and serum is lower than the model group, differences are statistically significant (P<0.01),the level of LD in the prostate tissue and serum compared with the model group is not statistically significant (P>0.05)2.ELISA testing β-EP express level:the level of P-EP in the prostate tissue and serum of the model group is low, compared with the normal group,differences are statistically significant (P<0.01),after medical treatment, the level of HD,MD,QLT in the prostate tissue and serum is higher than the model group,differences are statistically significant (P<0.01),the level of LD in the prostate tissue and serum compared with the model group is not statistically significant (P>0.05)3. ELISA testing IL-6express level in the prostate tissue:the level of IL-6in the prostate tissue and serum of the model group is low,compared with the normal group,differences are statistically significant (P<0.01), after medical treatment, the level of HD,QLT in the prostate tissue and serum is higher than the model group,differences are statistically significant (P<0.01),and any group of IL-6expressing level in serum is not statistically significant (P>0.05)4.RT-PCR testing IL-6,NGF level:the CT value of IL-6in the prostate tissue in the model group is on the rise,while the CT value of NGF is low compared with the normal group,differences are statistically significant(P<0.01);after medical treatment, the CT value of IL-6in the prostate tissue in the HD,QLT is lower than the model group,the CT value of NGF is high, compared with the model group, differences are statistically significant (P<0.01);the CT value of NGF, IL-6in the prostate tissue in LD compared with the model group is not statistically significant (P>0.05)Conclusion:1. Adopting pure rat prostate protein and complete Freund’s adjuvant produce homogenate,then using the homogenate injects the prostate combining Incomplete Freund’s adjuvant injecting under the skin is a successful way to improve the rat model.2.Danpu capsule can relieve CPPS, the mechanism is maybe that Danpu capsule inhibit the express of NGF and increase the express of IL-6,β-EP.3. The level of IL-6in serum is not statistically significant among any group,and the level of IL-6in the prostate tissue has the similar result with other factor resulting in pain,it can indicate that the change of IL-6in local tissue has not relationship with the change of IL-6in serum.and the impact of IL-6is maybe the result of local cell’secretion.4.After drug therapy,it can elevate the level of IL-6in tissue,which is similar with β-EP in tissue and serum,while the level of NGF decreases.so the degree of pain may has negative correlation with the level of IL-6, β-EP. and has positive correlation with NGF.5.DP capsule(HD) and QLT do not return the index to normal,the reason is maybe that the treatment cycle are not long enough.
