Objective:Hypoxia-inducible favtor (Hif-la) is the central mediator of cellular responses to low oxygen and vital to many aspects of cancer, In order to study celastol on the effects of the activion of hif-1α(hypoxia-inducible factor-1α)in human hepatoma cells activion, bring more helthy to the people. The expression level of (hypoxia-inducible factor-1α) hif-1α which in human cancerMethods:Hep3B、Hela, SK-Hepl was detected by Westren blot; MG-132and CHX are both kinds of proteasome inhibitor,we use them to find out celastol on the effects of hif-1α (hypoxia-inducible factor-la) by Westren blot; To observe Celastrol prevented hypoxia-induced expression of HIF-la target genes for vascular endothelial growth factor (VEGF) and EPO mRNA activion in activion by RT-PCR; The activity of Celastrol to inhibit hif-1α was determind by Hep3B-HRE cells, we used the Luciferase Reporter Assay; The activities of Hep3B proliferation cells were detected by MTT assay; To study Celastrol on the Hep3B invasion and cell viability we used the Transwell; To build the model of the BALB/c mice with human cancer Hela, when the tumor grown up,make them to3group,give the different concentrations celastrol to the nude mice stomach, measurement the volume of the tumor, record the date and make the tumor growth curve. Research celastrol of the function to nude mice tumor tissue on HIF-la by Westren blot.Results:Celastrol appear to dose-dependent inhibition of HIF-a protein activtition when the Hep3B cells treat with CoCI2and in hypoxia incubation by Western blot:Adopt MG132and CHX we find that Celastrol to HIF-a protein synthesis but not HIF-a protein degradation; Celastrol appear to dose-dependent inhibition of VEGF、EPO mRNA expression levels by PCR when the Hep3B cells in hypoxia incubation; we find that Celastrol have activation through Hep3B-HRE cells by Luciferase and MTT; Celastrol have a dose-dependent inhibit Hep3B cells transfer and invasion ability; Celastrol have the inhibition of the nude mice tumor and inhibit the HIF-a protein activtition which in the nude mice tumor tissue by Westren blot.Conclusion:Celastrol have the function of anti-tumor and HIF-α protein synthesis but not HIF-a protein degradation,Make contribution to the tumor therapy and prevention.
