BackgroundLiver fibrosis, a wound-healing response due to various reasons, is one of the most significant pathological manifestation of chronic liver disease,as well as an important step of liver cirrhosis. With the deep study of the cytobiology and molecular biolog, the traditional view about irreversible progress of chronic liver fibrosis has changed.lt had been confirmed decompensated cirrhosis were reversional, even the advanced cirrhosis. Hepatic fibrosis could be relieved by Chinese traditional medicine though multi-channel,multi-target, various cytokines and corss-talk of intracellular signaling passway,which drawed attention by academics and clinicians increasingly. Our previous studies have shown that the American cockroach extract has an postive anti-fibrosis effect of rat modle of hepatic fibrosis.Dose this effect could be replicate on other models and what is its mechanism,those the problems need to deeply study.objectiveTo observe the effects of Periplaneta americana extract sticky sugar amino acid treatment of rat liver fibrosis,and take the preliminary discussion on its resistance mechanism of action of liver fibrosis.Methods70male SD rats were randomly divided into7groups, namely the normal control group, model group, sticky sugar amino acid high dose group, sticky sugar amino acid in dose group, sticky sugar amino acid low dose group, notoginseng salvia miltiorrhiza and colchicine positive drug control group. In addition to the normal control group, the rest of the groups were respectively made die by administrated of intraperitoneal injection of pig serum.after7w, normal control group and model groups with physiological saline; sticky sugar amino acid group, notoginseng salvia miltiorrhiza group and colchicine group were daily irrigated stomach with suit corresponding subjects drugs, continues7w another.All rats were killed after the final lavage executed, routine tested liver function;meanwhile, line sapanwood element a eosin staining, Masson staining; Immunohistochemical were taken to detect liver organization transformation growth factor β1(TGF β1), organization metal protease inhibitor (TIMP-1); a-smooth muscle actin (a-SMA); nuclear transcription factors-κB (NF-κB). Radiation immune detection serum hyaluronic acid (HA), laminin (LN), type Ⅲ before collagen (P-C Ⅲ), Ⅳ collagen type (C Ⅳ). The comparison between the multiple data use SPSS17.0statistical software for single factor analysis of variance.Results1.The sticky sugar acid extracted from periplaneta americana could improve the rats, general condition such as spirit, activity, reactivity, fur, etc.The rats were suffered from liver fibrosis through immune response by Pig serum.Moreover, the medium and high dose drugs had better effect than other groups.2. After the rats were treated by the drug above.The specimens of their liver were better than model group in color, surface smoothness, edge, texture, swelling. And the3.The drug could reduce indexes of liver function including the serum alanine aminotransferase(ALT) and aspartate aminotransferase (AST) and so on in the rats which were suffered from liver fibrosis through immune response by drugs.The medium and high doses groups decreased more than the other groups, but there was no significant difference in medium and high dose groups; 4.The drug could reduce four indexes of hepatic fibrosis including the serum HA, LN, P-C Ⅲ, C Ⅳ. It could also alleviate pathological damage for liver fibrosis, reduce G, S pathological scores. The medium and high doses groups decreased more than the other groups, but there was no significant difference between medium and high dose groups;5.The drug could reduced the expression of TGFβ-1, TIMP-1, a-SMA and NF-κB in liver tissue of the rats which were suffered from liver fibrosis through immune response by drugs. The medium and high doses groups decreased more than the other groups, but there was no significant difference between medium and high dose groups.Conclusion1. The sticky sugar acid extracted from periplaneta americana could improve the rats, general condition such as spirit, activity, reactivity, fur, etc.The rats were suffered from liver fibrosis through immune response by drugs.Moreover, the medium and high dose drugs had better effect than other groups.2. The drug could alleviate pathological damage and improve liver function in rats which were suffered from liver fibrosis through immune response by drugs. The medium and high doses groups could improve their liver function more than the other groups;3. The drug could reverse hepatic fibrosis in rats which were suffered from liver fibrosis through immune response by drugs. Moreover, the effect of medium and high dose drugs groups is more obvious than other groups.4. The drug could keep the rats sufferring from liver fibrosis through immune response.It could improve liver function and reverse liver fibrosis pathology. The reason may be that it could reduce TGF-β1expression in the liver, inhibit HSC activation and proliferation and reduce TIMP-1levels.5. The drug could keep the rats sufferring from liver fibrosis through immune response.It could reverse liver fibrosis pathology for immune response by drugs.The mechanismmay be that it could relieve inflammationby reducing NF-κB protein expression andthen inhibiting the inflammatory cytokine interleukin-6(interleukin-6, IL-6), adhesion proteins such as ICAM-1expression; it could inhibit collagen gene expression in downstream NF-κB.
