[Objective]:Imitating pulmonary circulation and heart perfusion in isolated lung to explore the physiology of preservation methods,it can protect the activity,organization structure and normal function in isolated lung and provide quality pulmonary source for lung transplant patients.[Methods]:1. Using different methods to lavage the isolated lungs which from mongrel dogs,The methods (Cardiopulmonary bypass machine perfusion, Pressure perfusion, simple cryopreservation).The microscope is used to observe changes in tissue morphology and function of the lung tissue of different preservation methods and different time periods;2. Using immunohistochemical detection to detect the activity of NF-KB in macrophages in the lung tissue by different preservation methods and in different time periods;3. Using microscopic to observe the HE biopsy of lung tissue.To count the number of the macrophages by immunohistochemistry Semi-quantitative integration method and positive degree of NF-KB;[Results]:1. We Successfully set up a isolated lung protection model of dog by three different protections methods to save isolated lung (Cardiopulmonary bypass machine perfusion group, Pressure perfusion group, simple cryopreservation group). 2. Finding Cardiopulmonary bypass machine perfusion group has complete structure of alveolar tissue, fewer inflammatory cells, little, exudate, spacing slightly widened by observing the HE biopsy to compare the three groups,.Pressure perfusion group is followed, simple cryopreservation group is poor., With the growth of time away from the body comparison within groups, The alveolar tissue structure gradually collapse, inflammatory cells and exudate increased.The alveolar spacing gradually widened, alveolar space visible more exudate.3. Finding Cardiopulmonary bypass machine perfusion group has fewer macrophages by observing pathological immune staining the biopsy to compare three groups, NF-KB positive degree is lower, the macrophages in Pressure perfusion groups are slightly more, NF-KB positive degree is higher.The macrophages in simple cryopreservation group are more,.NF-KB positive degree is strong. With the growth of time away from the body, an increase in the number of macrophages in the three groups and enhance the level of NF-KB positive.[Conclusion]:1. The Cardiopulmonary bypass machine perfusion can protect the isolated lung tissue better and retain its structural and functional integrity,pressure perfusion is followed,the simple cryopreservation is worse than the other methods.With the growth of time away from the body, the lung tissue structure and function become worse;2. The inflammatory response of Cardiopulmonary bypass machine perfusion group is lighter, the macrophages are Seldom.the pressure perfusion group is slightly stronger, the simple cryopreservation group is slightly seriousr; with the growth of time away from the body, an increase in the number of macrophages enhanced activity.
