Major depressive disorder is a mental disorder characterized by depressed moodand anhedonia. MDD has the high rates in the global scope, which costs a lot because ofthe disabling disease even lead to suicide. The present studies show that multiplebiological systems are associated with the onset of depression, but the treatment ofdepression medication is basically depends on the role of monoamine neurotransmittersystem. However, numerous studies have reported that the drugs in the treatment ofpatients with depression not only has low remission, but also has side effects whichcannot be ignored, such as calming effect, sleep disorder and cognitive impairment, thusdeveloping new consideration with little limitation and more safe antidepressants isimminent.This study will research the antidepressant effects of Salvianolic acid B (SalB)which was gained after extraction, separation and purification from salvia miltiorrhiza.By tail suspension and forced swimming models, classic antidepressant drug evaluationmodels to evaluate the acute antidepressant effect of SalB. Through chronic mild stressdepression model evaluate the long-term efficacy of SalB and study its pharmacologicalmechanism.The experimental results show that SalB owns obvious antidepressant effects inanimal experiments. In acute experimental tail suspension and forced swimmingexperiment, SalB significantly reduced the proportion of the immobility. In comparisonwith imipramine (as for reference drug), we found that SalB won’t reduce the autonomicactivities in mice, explaining it plays to the strong antidepressant role and would notinduce sedation effect at the same time and declaring SalB has antidepressants with lessside effects. In this simulation, the chronic mild stress(CMS) in animal models, SalBcan effectively relieve depressive symptoms of CMS, and has the ability to restore theincrease caused by the number of corticosterone(CORT),explaining the antidepressanteffects of SalB may influence the HPA axis system. This thesis further showed that theantidepressant mechanism of SalB was inhibiting the pro-inflammatory cytokines IL-1β and TNF-α in cortex. This paper provides a new choice for depression treatment, fromthe behavior and molecular experiments, we confirmed that the antidepressant effects ofSalB, and SalB acts through the anti-inflammatory efficacy.
