Objective:This paper focuses on the protective effects and mechanism of Buchang Naoxintong capsule combined with Danhong injection in the treatment of acute cerebral ischemia reperfusion injury, so as to provide theoretical and experimental basis for further study on clinical effects and usage of Buchang Naoxintong capsule and Danhong injection against cerebral ischemia.Methods:With the help of Zea Longa modified thread embolism method, transient middle cerebral artery occlusion model of rats is prepared, containing sham operation group, MCAO group and Naoxintong group, Danhong group and Combined group, all of which are divided into three subgroups,1d,3d and7d. After reperfusion, the animals are killed respectively on the1st day,3day and7th day, and then their brain tissues are rapidly taken to have experiments, each corresponding time point respectively at Id,3d and7d, were:1) Neurological Severity Score (NSS) is used to evaluate the neural function;2) TTC-dying method to measure the cerebral infarction volume;3) Dry and wet weight method to measure the brain water content;4) EB method to measure the cerebral capillary permeability;5) TUNEL method to detect cell apoptosis6) Western blotting to detect the protein expressions of B1R and B2R in ischemic brain tissue Results:1) Compared with the sham operation group, the neurological severity score, infarction volume, brain water content, the cerebral capillary permeability and the number of apoptotic cells around the infarction of rats in MCAO group, Naoxintong group, Danhong group and Combined group all increase significantly (P<0.05).2) Compared with the MCAO group, the neurological severity score, infarction volume, brain water content, cerebral capillary permeability and the number of apoptotic cells around the infarction of rats in MCAO group, Naoxintong group, Danhong group and Combined group all decrease significantly (P<0.05). The effects of Naoxintong capsule combined with Danhong injection in treatment is better than that of respective Naoxintong capsules or Danhong injection (P<0.05).3) On the3rd day after ischemia reperfusion, Naoxintong capsules can effectively decrease the expression of B1R (P<0.05), the expression of B2R had no significant difference (P>0.05); Danhong injection can reduce the expression of B2R (P<0.05), but no significant differences occurs to B1R expression (P>0.05). The effects of Naoxintong capsule combined with Danhong injection intervention can reduce MCAO rats’expression in B1R and B2R (P<0.05).Conclusions:1) Naoxintong capsule combined with Danhong injection treatment can reduce the score of severity of MCAO rats’nervous system disease, cerebral infarction volume, brain water content, the cerebral capillary permeability around the infarction and the number of apoptotic cells,also can hinder MCAO rats’expression ability in B2R and B1R.2) Naoxintong capsule combined with Danhong injection can protect the acute brain ischemia reperfusion tissues, is better than that of respective Naoxintong capsules or Danhong injection in treatment.
