ObjectiveThe purpose of this experimental study is exploring the role of the cytochrome C oxidase (cytochrome c oxidase5A, COX5A) in the spinal neuroplasticity by using Partial removal of animal dorsal root ganglion (L3-L4,L6were ganglonectomy, L5was spared) in Sprague-Dawley (SD) rats and electric acupuncture rats.MethodsHealthy adult Sprague-Dawley (SD) rats were randomly divided into:normal group, the surgical group, Partial ganglonectomy with sciatic nerve injection COX5A interference group, Partial ganglonectomy with sciatic nerve injection empty vector group. Each group consists of five rats. The surgery group only removed the rats dorsal root ganglion bilateral L3, L4, L6, spared bilateral L5dorsal root ganglion. COX5A interference gene fragments HIV lentiviral expression vector was injected into the sciatic nerve to block cox5A in dorsal root ganglion. Empty vector injection was as negative control. All rats were routinely given drinking water, feed and timely replacement of litter, do not do any special treatment. Rats is each group were peformed pressure claws and tail flick pain and temperature detectionwithin7days,14days,28days, respectively, then sacrificed at28days for the fluorescence histochemical staining for CGRP, and the number CGRP positive varicosities in lamina II was counted, then experienced statistic analysis. Results1.COX5A interference expression vector was successfully constructed. Sequencing results confirmed that the target sequence was in accordance with the stand sequence checked up in CNBI. Construct a viral vector to the success of the package out of the pseudo-viral particles, successfully transfected into PC12cells.2.The pressure the claw reflection results display partial removal of the dorsal root ganglion surgery leads to Rat Paw Pressure stimulus time (P<0.05) significantly delayed, whereas electro-acupuncture (EA) delayed pressure feeling significantly, also (P<0.05). Furthermore, COX5A interference prolonged significantly the parameter in EA rats, when compared with empty vector EA rats (P<0.05). This suggested that COX5A interference is effect in the recovery of sensory functiony.3. The tail flick measurement showed the similar changes to pressure the claw reflection. However, there is no statistic significance among groups.4. At28days after the dorsal root ganglionectomy, number of CGRP in Lamina Ⅱ of the spinal cord significantly reduced than noramal one(P<0.05). EA could reverse this reduction and give significan difference (P<0.05). Moreover, COX5A interference decreased significantly the number of CGRP in Lamina Ⅱ in EA rats, than empty vector rats with EA.(P<0.05).Conclusions1. The lentiviral vector COX5A interference RNA is successfully constructed.2. By the partial removal of dorsal root ganglion, we proved in rats with partial doral root defferentaion, pain and temperature changes is associated with the expression of COX5A, suggesting the role of COX5A in neuroplasticity in rats with EA. These findings could address that cox5A treatment and mechanism of EA in the neuroplasticity.
