| [Objective] Research the part of mitochondrial DNA sequences of Schistosoma japonicum in Dali, Yunnan province, and explore the relationship of Schistosoma japonicum between different schistosomas by genetic structure of species. And lay a foundation for the learning and understanding of the biological characteristics of Schistosoma japonicum in the future.[Methods] The cercariaeums of Schistosoma japonicum were obtained from masculine oncomelania hupensis from Majiuyi. Then every Kunming mouse is infected with20~30cercatiae of Schisosoma japonicum via abdominal skin.Mice were killed on the45th day after infection. Schistosome japonicum adults were collected from mice through perfusion method. The genomic DNA was extracted with DNA extraction kit, and the complete gene fragments of the mitochondrial CO1, Cytb, ND1, ND6, ND4L were amplified by PCR and sequenced by biotech company. Make some gene bioinformatic analysis for these sequences by bioinformatic softwares. Make a phylogenetic tree of schistosome mitochondrial CO1, Cytb, ND1, ND6, ND4L with the same sequences of other schistosomes which searched in GeneBank by DNAstar and Mega4.0in order to analyze the relationship between species in gene level.[Results] The mitochondrial CO1, Cytb, ND1, ND6, and ND4L gene fragments of Schistosoma japonicum in Majiuyi are obtained. The phylogenetic tree show significant branch when these seqences compares to the same sequences of S.mansoni, S.haematobuim, S.mekongi, S.malayensis. However, it can’t constitute a separate branch when compare to the other Schistosoma japonicum mitochondrial sequences which is already published in Genebank. The results indicate that the samples of this experiment is attributed to chinese Schistosoma japonicum strains.[Conclusion] The mitochondrial CO1Cytb ND1ND6and ND4L gene fragments of Schistosoma japonicum in Dali, Yunnan Province are obtained. The results of phylogenetic tree mean that the Schistosoma japonicum of Dali attribute to the group of the chinese Schistosoma japonicum. And the mtDNA sequences of Schistosoma japonicum has significant branch in phylogenetic tree compare to S.mansoni, S.haematobuim, S.mekongi, S.malayensis. |
