ObjectiveWith the continuous development of the social economy and the improvement ofthe quality of modern life, obesity has become a global social problem. However, it isvery clear that how difficult to lose weight, and diet pills have limited effect andmultiple side effects, therefore, designing a new, effective and low side effects weightloss method has become more and more imperative. This study is trying to choose anodorant, using it for the weight regulation through olfactory pathway from olfactoryepithelium to the hypothalamus by sniffing it.MethodsAfter viewing a few relevant papers, grapefruit essential oil (GEO) has becomethe one chosen for this study. First, using GC-MS for the analysis of composition ofGEO in the concentration of2%and5%(dissolved in N-hexane).Second, dividing72adult male C57BL/6mice into3groups, which were blank group(BG), high caloriegroup(HCG) and DXM group(DG).BG and DG was using a regular diet, HCG wasusing a high glucose and lipid diet. DG was injected DXM into the intraperitonealevery72h. All groups was treated like this for60d, with keeping records of weight ofeach group before/after modeling. Third, due to HCG became a failed modeling group,DG was the only group for the next sniffing experiment. DG was divided into2groups, which were sniffing blank group(SBG) and sniffing essential oilgroup(SEOG). SBG was sniffing air, and SEOG was sniffing grapefruit essential oileveryday for30d, with records of weight of each group before/after sniffing. Results1. In the concentration of2%, there was only one compound was discovered, it was(R)-cyclohexene,1-methyl-5-(1-methyllethenyl);in the concentration of5%, therewere7compounds were discovered, they were (1)(R)-cyclohexene,1-methyl-5-(1-methyllethenyl)(D-limonene)(2)1,6-octadien-3-ol,3,7-dimethyl (3) trans-p-mentha-2,8-dienol (4) cis-limoneneoxide (5) trans-limonene oxide (6)(R)-3-hydroxybutyric acid (7)(R)-2-cyclohexen-1-one-2-methyl-5-(1-methylethenyl). D-limonene is the majorcomposition of the oil, reaching the level of97.56%.2. With a comparision to BG, the body weight of HCG has droped instead of rising,which indicated that a modeling failure. But, the body weight of DG showedrisen(α<0.05),which indicated a successful modeling group.3. The body weight of SBG was still going up for the next30days, but SEOG wasgoing down(α<0.05), which means sniffing grapefruit essential is effective forthe body weight regulation.Conclusion1. There may be effective substances in GEO that can regulate the weight changes,in this case, it may be D-limonene.2. DXM injection into intraperitoneal may make a obese model.3. Through sniffing grapefruit essential oil, the body weight of obese mice decreased.There may be a new way for controlling appetite and losing weight.
