Objective: Investigate siRNA silence survivin gene expression in nasopharyngealcarcinoma (NPC)cells cycle, value-added, apoptosis and Caspase3, the influence of theenzyme activity, provide new experimental basis for nasopharyngeal carcinoma genetherapy.Method: Design and synthesis of two different specific siRNA interference survivinexpression, by liposome transfection nasopharyngeal carcinoma CNE2cells. Using flowcytometry instrument, detection of cell cycle and cell apoptosis rate; Using CCK8reagentmethod, detection cell value; Using Caspase3kits, detection of Caspase3enzyme activity.Results: After siRNA silences Survivin gene expression, compared with blank controlgroup, negative control group, Survivin siRNA in group a, Survivin siRNA b group ofcells in G0/G1phase, the G1phase of the cell increased, S phase cells reduce (p﹤0.05);Compared with blank control group, Survivin siRNA in group a, Survivin siRNA group bcell proliferation was inhibited (p﹤0.05), Survivin siRNA group b and Survivin siRNAcompared group a, cell proliferation inhibition significantly more (p﹤0.05); Survivinapoptosis rate of siRNA group b (39.5%±0.79) higher than the blank control group(2.65%±0.41), the liposome group (3.25%±0.52), negative control group (4.16%±0.62) and Survivin siRNA in group a (37.2%±0.71)(p﹤0.05); Caspase3activitydetection showed that compared with blank control group, negative control group, SurvivinsiRNA in group a, Survivin siRNA Caspase3enzyme activity of group b (p﹤0.05).Conclusion:1. Survivin gene expression after siRNA silence, inhibits nasopharyngealcarcinoma CNE2cells growth, promoting apoptosis, in G0/G1phase retardation.2. Survivin gene expression after siRNA silence, Caspase-3innasopharyngeal carcinoma CNE2cells enhanced enzyme activity. 3.
