Objective: To investigate the value and expression of cannabinoid receptor1(CB1) and focal adhesion kinase(FAK) in the hepatic tissue of mice withschistosome’s liver fibrosis,so as to explore the further mechanisms of schistosoma offibrosis.Methods:40healthy Kunming mice were randomly divided into two subgroups:normal group(10mice) and model group(30mice). Mice of model group were infectedsubcutaneously with cercariae of schistosoma japonicum. Mice were raised for8weeksnormally, killed for liver tissue. Then the model group were devidede into three groupaccording to the fibrosis level of liver tissue. The pathological change was tested by HEstaining, fibrosis level was observed by Masson staining,The expression of CB1of eachgroup in liver tissues were observed by immune histechemical technique,the expression ofFAKmRNA and CB1mRNA were tested by reverse transcription polymerase chainreaction(RT-PCR).Results:1、8weeks later, HE staining and Masson staining showed that the structure in thenormal liver was natural and the one of model group was destroyed, Schistosoma eggsdeposited in the liver tissue, there were abundant fibrous connective tissue and inflammatorycells around the worm tubercles, which supposed that mice liver fibrosis of schistosoma wasformed and the model was successful.2、Compared with the normal group, there were worm tubercles and pathologicalchanges in liver of model group(P<0.01); The expression of CB1、CB1mRNA and FAKmRNA could be observed around worm tubercles and portal vein in tested group(P<0.01). 3、The expression of CB1、CB1mRNA and FAKmRNA could be seen obviously intested group and gradually became more intensive along with liver fibrosis increasing (P<0.05或<0.01)Conclusions:1、CB1involved in the formation and development of schistosoma liver fibrosis.2、CB1may promote progression of schistosoma liver fibrosis by activating FAK,then activating PI3K.
