Objective: Study on the change and significance of hypoxia-inducible factor-1α(HIF-1α), adrenomedullin (ADM),endothelin-1(ET-1) levels in the neonatal rats as theywere in the period hypoxia-induced pulmonary hypertension (HPH). Methods:120newborn Wistar rats were randomly divided into hypoxia group and the oxygen control,newborn rat HPH model is set up, respectively in the anoxic3d,5d,7d,10d,14d and21d take hypoxia group and the control group on the same day of age determination ofnewborn rat every10average pulmonary artery pressure, serum HIF-1alpha, ADM andET1levels.Results:(1) Hypoxia group of newborn rat serum mean pulmonary arteriapressure in hypoxia in3d,5d,7d,10d,14d,21d is significantly higher than the controlgroup, the difference was statistically significant (P <0.05).(2) The rats in hypoxia grouphad a higher serum HIF-1α than the controlled group as they experienced hypoxia of3,5,7,10and14days (P<0.05).(3) Serum content of ADM was significantly higher than thecontrol group (P<0.05) after a7-day hypoxia, but there was no significant difference aftera hypoxia of10,14or21days, compared with the control group (P>0.05),(4) Serum ET-1levels were clearly higher than the control group (P<0.01).(5) Elevated serum alpha HIF-1with increased mean pulmonary arteria pressure positive correlation (r=0.279, P <0.01), serum levels of ET-1increases with increased mean pulmonary arteria pressurepositive correlation (r=0.341, P <0.01), serum ADM content change with increased meanpulmonary arteria pressure no correlation (r=0.127, P>0.127).(6) Serum levels of ET1levels rise with HIF-1alpha content higher positive correlation (r=0.561, P <0.01),plasma ADM levels rise with HIF-1alpha content higher positive correlation (r=0.348,P <0.01), the changes in concentrations of ADM and ET-1no correlation (r=0.157, P> 0.157).Conclusion:1)As a key factor, HIF-1α made ET-1and ADM expression raised inthe pathogenesis of hypoxia-induced pulmonary hypertension (HPH) of the neonatal ratsand caused a unbalance of ET-1and ADM. Thus HIF-1α played an important role inneonatal rats.2) HIF-1α, ADM and ET-1altogether contributed to the occurrence anddevelopment of HPH in neonatal rats..
