Objective:To observe the effects and mechanism of YiQifusheng decoction on intestinal mucosal barrier in tumor-bearing experimental mice with gastric cancer induced by inoculateing tumor blocks.Method:Tumor blocks were inoculated into t he Kunming mice subcutaneously to form the gastric cancer entity-tumor mock-up. Fifty young Kunming mice were randomly assigned into normal group, model grou p、CHM group(YiQifusheng decoction group)、WM group(5-Fluorouracil group)、co mbination group(YiQifusheng decoction and5-Fluorouracil group),10mice in each group.The CHM group was perfused stomach with YiQifusheng decoction0.5ml on ce a day caccording to the recommended dosage(23.3g/kg,0.5ml/day), toally14da ys. The WM group was injected into mice abdomen with0.2ml5-fluorouracil once a day(caccording to20mg/kg), toally5days. The combination group was perfuse d stomach with0.5ml YiQifusheng decoction once a day caccording to the recomm ended dosage(23.3g/kg,0.5ml/day), toally14days, and meanwhile injected into mi ce abdomen with0.2ml5-fluorouracil once a day(acccording to20mg/kg), toally5days. After14days, to kill the mouse and get the tumor and the bowels in the same position,weight the tumor up,then to count the inhibiting tumor ratio.INR=(the average weight of the model group-the average weight of the experimental group)/the average weight of the model group×100%.D-Lactate、diamine oxidase、 endotoxin and Immunoglobulin G were measured by Enzyme-linked immun the sosorbent (ELISA)assay.The pathological changes of the bowels iname position was observed by light microscopy after Hematoxylin Eosin(HE) staining.After that, SA BC immunohistochemistry was used to examine the expression of intestinal tightjun ction zonula occludens-1(ZO-1)protein.ResuIts:1.Comparing with the normal group, the levels of serum D-Lactate、diamine oxidase、endotoxin and Immunoglobulin G in the model group increased higher significantly(P<0.01), the levels of ZO-1decr eased significantly(P<0.05).2. Comparing with the model group, the levels of serum D-Lactate、diamine oxidase、endotoxin and Immunoglobulin G in the CHM grou p、the WM group、the combination group decreased higher significantly(P<0.01), t he levels of ZO-1increased significantly(P<0.05).3. Comparing with the normal gro up, the levels of serum D-Lactate、diamine oxidase、endotoxin and Immunoglobul in G in the combination group is more close to the the normal group.4. Comparin g with the normal group,the inhibiting tumor ratio of the combination group increa sed higher significantly(P<0.01), the inhibiting tumor ratio of the CHM group and the WM group increased significantly(P<0.05).5. In the normal group, complete in testinal villus structure was observed by light microscopy. In the model group, spar se intestinal villi and structural damage were observed. In the CHM group and the WM group, pathological changes of intestinal mucosa were reduced than model group.ConcIusion:1.YiQifusheng decoction can recover the integrity of intestinal mu cosal barrier in tumor-bearing experimental rice with gastric cancer and inhbit the growth of the tumor.;2.YiQifusheng decoction can recover the integrity of intestina1mucosal barrier maybe by decreasing the levels of serum D-Lactate、diamine oxi dase、endotoxin and immunoglobulin G, increasing the expression of intestinal tight junction zonula occludens-1(ZO-1)protein in the mainwhile;3.YiQifusheng decoctio n has reasonable structure, legislative theory and practical basis.It has appropriate e fficacy and effection of recovery to the integrity of intestinal mucosal barrier in tu mor-bearing experimental rice with gastric cancer.
