Objective: To research the Single Nucleotide Polymorphism (SNP) of solute carrierfamily19(SLC19A1) in the group of55acute lymphoblastic leukemia(ALL) patients andmalignant lymphoma (ML) between the December in2011and January in2013in theFirst Affiliated Hospital of Xinjiang Medical University (XJMU).To analyze thedistribution of several genotype in different racial groups and to investigate the relevancebetween the genotype and adverse effects on chemotherapy by high dose Methotrexate. Inorder to provide the reference for individual treatment in MTX. Methods:1. Potassiumiodide method and kit assay was used to study the DNA by blood samples respectively.Polymerase chain reaction-Restriction fragment length polymorphism (PCR-RFLP)detects the polymorphism of SLC19A1A80G and analyzes the distribution of genotype indifferent racial group.2. enzyme multiplied immunoassay technique was utilized to testblood concentration.3. Case history and information of the patients were collected toobserve the incidence of adverse effect during the chemotherapy and to analyze therelations with each genotype by the comparison the laboratory indexing of HD-MTXbefore and after chemotherapy in different genotype. Results:1. The purity of DNAextracted by potassium iodide method and kit assay shows significant difference (P<0.05).2. There is no significant difference in the diversity of genotype between Han and Uyghurin both male and female in our research(P>0.05).3. In our study, the number ofPlatelet(PLT) in change shows the significant difference between the group of GG and AA(P<0.05).4. Compared with the group of AG and GG, the genotype of AA express thesignificant difference on the change of alanine aminotransferase (ALT) after HD-MTXchemotherapy (P<0.05), and for the comparison to AG the direct bilirubin in the genotypeof AA and GG express the significant difference (P<0.05).5. In our study, there is the significant difference of the genotype distribution frequency by SLC19A1in hepaticinjury patients (P<0.05). Conclusion:1. In our study, the distribution frequency of thegenotype in SLC19A1shows no significant difference in the group of Chinese Han andUyghur, and there is also no significant difference of the distribution frequency of thegenotype between male and female patients.2. The results of our study showed thepatients with genotype of GG in the deacrease of Platelet(PLT) is witnessed more obviousthan that of the genotype in AA. Furthermore,the patients with genotype of AG and GG inSLC19A1show a remarkable increase of aminotransferase than the figure for AA, and thedirect bilirubin increases most in the genotype of AG than the data for other two genotypes.3.The polymorphism of SLC19A1may remarkably associate with the hepatic injurypatients during the HD-MTX chemotherapy.
