Objective: Based on previous study on the SNPs of PLCE1, we investigate the expression ofPLCE1gene mRNA and protein in Kazakh’s esophagus squamous cell carcinoma (ESCC), as wellas to evaluate association between PLCE1expression and clinicopathologic characteristics ofESCC.Methods: qRT-PCR was employed to detect the expression of PLCE1mRNA in50paraffin-embedded Kazakh ESCCs and50non-cancerous adjacent tissues, and staining of tissuemicroarray were respectively conducted to detect the expression of PLCE1protein in110paraffin-embedded Kazakh ESCCs, and50non-cancerous adjacent tissues.Results:1. The expression of PLCE1mRNA in ESCC was significant higher than that innormal-appearing mucosa(P=0.012); the expression of PLCE1mRNA was associated withregional lymph node metastasis(P=0.001) and pathological staging(P=0.022);the expression ofPLCE1mRNA had no relationship with the degree of differentiation、gender and age (P>0.05).2.PLCE1mainly located in cytoplasm. The positive-expression rate of PLCE1was79.1%(87/110)in ESCC and38%(19/50) in non-cancerous adjacent tissue, the difference was significant(P<0.05). Expression of PLCE1protein was significantly correlated with lymph node metastasis(73.2%in N0cases,89.7%%in N1cases, P=0.042) and pathological stage (71.9%in stage I–IIcases,89.1%in stage III-IV cases, P=0.028), respectively. In contrast, no statistically significantcorrelations with gender (P=0.052), age (P=0.366) and histological grade (P=0.500) weredemonstrated for expression of PLCE1protein.Conclusions: The over-expression of PLCE1mRNA and protein was related to the genesis ordevelopment or metastasis of ESCC and might be a useful parameter as an indication of tumorinvasion, metastasis and biological behavior of human ESCC. |
