Purpose:Study the effect of follicular fluid of patients with endometriosis on vitro human ovarian endometrial luteinized granulosa cell proliferation and secretion of estradiol (E2),progesterone (P), and the result after FSH and of BMP-15functions.Materials and methods:Take cumulus granulosa cells around oocyte maturing on the same day after ovulation COH from infertility IVF-ET patients due to tube factor/male factor, and purify, divide the cells into two groups:control group and experimental group. Control group is cultured with DMEM/F-12nutrient solution while experimental group were cultured with DMEM/F-12nutrient solution added with5%tube factor/male factor infertility patients follicular fluid(tube/male group),5%endometriosis infertility patients follicular fluid(endometriosis group),5%endometriosis infertility patients follicular fluid+12ng/ml FSH(endometriosis+FSH group),5%endometriosis infertility patients follicular fluid+6ng/mlBMP-15(endometriosis+BMP-15group),respectively. After culturing for48h,calculate the amount of granulosa cell using cytometry. Meanwhile,collect the supernatant fluid and put it in-80℃refrigerator. Detect the E2and P value of the supernatant fluid using chemiluminescence method.Results:(1)After culturing for48h, the amount of granular cells in endometriosis group and endometriosis+BMP-15group are obviously less than control group, while endometriosis+FSH group, tube/male group show no obvious difference comparing with control group.(2) After culturing for48h, the control group and alternative experimental group show no special difference in E2secretion without considering the effect of the original follicular fluid. P secretion, the value of endometriosis group is lower than tube group(p<0.01), improved when adding FSH(p<0.05), but BPM-15can not change the result.Conclusion:Follicular fluid of endometriosis patients has suppression effect on human ovarian granulose cell proliferation and P secretion. FSH could improve this, while BMP-15shows no obvious effect. All have no obvious effect on E2secretion.
