Tripterygium Wilfordii Hook.f.(TWHF) is a traditional Chinese medicinal herb, whichhas a long using history in China. Its extracts have unexpected therapeutic effect onantiinflammatory, antitumor, antifertility and autoimmune diseases, such as systemic lupuserythematosus, rheumatoid arthritis, chronic nephritis, autoimmune disorders and so on. Itis the first choice on the clinical treatment of autoimmune diseases in China. However, it’stoxic effects and clinial side effects have to be concerned. Therefor, this thesis reports thatthe effective fractions of TWHF was extracted by using alkaline water method, and theTWHF-loaded nanostructured lipid carriers(TWHF-NLC) was prepared by using hot highpressure homogenization metod and its effects on Mice With IgA Nephropathy wasobserved. The main methods and results are as follows:(1)The effective fractions was washed by using5%sodium carbonate solution, it’s amixture mainly contained diterpenes and alkaloids. The content of the two parts wasmeasured by using UV spectrophotometry, the results are1.26%and75.61%. Triptolideand wilforine, which are the representative compounds of diterpenes and alkaloids inTWHF, are determined by using high efficiency liquid chromatography method. Thecontent of triptolide is0.054%, and wilforine is3.2%.(2)The TWHF-loaded nanostructured lipid carriers was prepared by using hot highpressure homogenization method. The optimized formulations of TWHF-NLC are:0.4%effective fractions of TWHF,3.2%glyceryl behenate(ATO888),0.8%propyleneglycol monocaprylate(C90),3.0%ethoxylated hydrogenated castor oil(RH40) and92.6%water. The preparetion pareameters were mix the oil phase and water phase at80℃,then high stirring at13000rpm for2min and high pressure microfluidics at800bar for3cycles. The size of TWHF-NLC was101.9±0.80nm and PDI is0.099±0.006. Theencapsulation efficiency of triptolide is46.57%, which was determined by ultrafiltrationmethod. And the encapsulation efficiency of wilforine is88.9%, and84.87%if the dextrangel column chromatography method was used. Transmission Electron Microscopydemonstrated that the particle size distributed in a small range.(3)Animal experiment demonstrated that TWHF-NLC performed obvious curative effect on mice wtih IgA Nephropathy. The kidney histopathology showed thatTWHF-NLC could reduce the collagen content among kidney cells, and could clear theMCP-1, restrainting its expressions in the meanwhile.
