AIM:To investigate the effects of the metabolites from hypoxia of cardiacmyocyte on migration and induction of rat cardiac fibroblastMETHODS:Cardiac myocytes and cardiac fibroblasts were isolated from theventricles of neonatal Sprague—Dawley rats and primarily cultured with digestion anddifferential adhesion.Cardiac myocytes used for hypoxia experimens for2hã€4hã€6hã€8hã€10hã€12h in the there days. Cardiac fibroblasts in3passages were used forTranswell chamber assay to determine the chemotatic effects of the effects of themetabolites from hypoxia of cardiac myocytes on chemotaxis.RESULTS:Compared with control group,the metabolites from hypoxia ofcardiac myocytes on migration of cardiac fibroblast that migrated through thepolycarbonate membrane were obviously increased in hypoxia for2hã€4hã€6hã€groups.The most migration was induced by the metabolites from the cardiac myocyteshypoxia for2h. however the migrate of rat cardiac fibroblast obviously wasinhibited after hypoxia for8h. The number of migrated cells in hypoxia for12h groupWas the lowest.CONCLUSION:(1) Early metabolites from the cardiac myocytes of hypoxia caninduce migration of cardiac fibroblast, it is the Initially factors that the cardiac fibroblastmigrate, Constitute the occurrence of cadiac fibrosis of Initially start-factor.(2) The metabolites of changed cardiac myocytes metabolic function had activatly repairedmechanism.The metabolites are induce migration of cardiac fibroblast to diseasedregion,the metabolites of inducing migration may not be related to inflammatory factor.(3) The metabolites from hypoxia of cardiac myocyte induce the migration of cardiacfibroblast, may indicate that the function of cardiac had damaged,It is the reason thatthe acute or chronic regeneration of repaired activate. |
