Purpose By using the immunofluorescence technology, we aimed at detecting theexpression and distribution characteristics of Beta2adrenergic receptor in theinfantilehemangiomas tissue and exploring its role and significance in the pathologyevolution of infantilehemangiomas.Methods HE staining to48cases of infantile hemangioma samples, according to theclinical and histologic features, they were divided into proliferating IH group andnon-proliferating IH group.29cases were in the proliferation period, while the other19cases were in the non-proliferation period. By using immunofluorescence technology, theendothelial nuclei and beta2adrenergic receptor of the IH tissue were marked byfluorescent tags respectively in two groups. Observe the location of fluorescent labelingshown in photos. Convert these fluorescent Images to black and white pictures by Image-Pro Plus6.0software firstly, and then analyze each photo. Calculate the average opticaldensity value of beta2adrenergic receptor in each case.Results The mean optical density value of endothelial cell core in Proliferating IH groupwas0.031±0.002, while this value in fading period IH group was0.027±0.002. There wassignificant statistical difference between the two groups. The mean optical density value ofbeta2adrenergic receptor Proliferating IH group was0.034±0.003while this value infading period IH group was0.027±0.002. There was significant statistical differencebetween the two groups. There was higher expression of beta2adrenergic receptor in the proliferating IH group than that of non-proliferating IH group. Beta2adrenergic receptorswere widely expressed in IH tissue, especially in endothelial cell nucleus showninfluorescent images.Conclusion There was significant higher expression of endothelial cell core and beta2adrenergic receptor in the proliferating IH group than those of fading period IH group. Asthe fade of endothelial cell core, the beta2adrenergic receptor expressed less in the IHtissue. We speculated that the beta2receptors are widely expressed in infantilehemangioma endothelial cells, and may exist in the rough cells. This pointed out that themechanism of beta blockers2agent applied to infantile hemangioma, inducing its fadingmay be related to beta2adrenergic receptors in endothelial cells and as well as rough cells.
