Objective:To study the expression of hyaluronan acid synthase family(HAS1HAS2、HAS3) in renal carcinoma and discuss the potential clinical significance.Methods:qPCR was applied to detect the expression of HAS1mRNA、HAS2mRNA and HAS3mRNA in five ccRCC cell lines (ACHN、CAKI-I、OS-RC-2、786-0、 SN12PM6) and normal human renal proximal tubular cell line(HK-2cells). Additionally, Western blot and immunohistochemical methods were taken to measure the expression of the HASs(HAS1、HAS2、HAS3) protein in above renal cancer cells and ccRCC tissues.Results:HAS2mRNA expression was obviously higher in the five ccRCC cell lines than that in the HK-2cells,especially in the SN12PM6cells (P<0.05). HAS1mRNA expression reduced distinctly in the five ccRCC cell lines compared with the HK-2cells(P<0.05). The expression of HAS3mRNA was also lower in the renal cancer cell lines than that in the HK-2cells except CAKI-I cells(P<0.05). The expression of HAS2protein significantly increased in the five ccRCC cell lines (P<0.05)whereas HAS1and HAS3protein had no obvious expression. In addition,the expression of HAS2protein was significant higher in the ccRCC tissues than those in the normal tissues, the positive percentage were85%(17/20)、20%(4/20) respectively, The difference was statistically significant (P<0.005),and its expression was related to tumor grading and stage, however the positive expression percentage of HAS1in the ccRCC tissues and the normal tissues were30%(6/20)、40%(8/20), respectively. The positive percentage of HAS3were35%(7/20)、20%(4/20), respectively. both HAS1and HAS3protein expression between the ccRCC tissues and the normal tissues have no statistically significant difference(P>0.05).Conclusions:Our study showes that expression of hyaluronan acid synthase family(HAS1、HAS2、HAS3) are different. HAS2is detected to highly express in the five ccRCC cell lines and ccRCC tissues whereas HAS1and HAS3have lower expression, which shows that HAS1and HAS3may play little role in renal carcinoma. On the contrary, HAS2may play an important role in the occurrence and develepment of renal carcinoma as a advantage subtype, and may serve as a potential molecular marker and potential therapeutic target for renal carcinoma.
