In this paper, the chondrocytes of rats were used as research materials, usingimmunohistochemistry, ELISA, RT-PCR, Western blot method, exploring theprotective effect and mechanism of total secoiridoid glucoside and gentiopicroside onchondrocytes induced by IL-1β; structures of compounds were identified by HPLC-MS.The study explore its mechanism on the basis of molecular level for the first time,providing theoretical basis for further development and resource substitution ofGentiana officinalis H.Smith, lay the foundation for finding the new anti-inflammatory drugs.Using immunohistochemistry, ELISA, RT-PCR, Western blot method, exploringthe protective effect and mechanism of total secoiridoid glucoside and gentiopicrosideon chondrocytes induced by IL-1β. The results shoeed that gentiopicroside and totalSecoiridoid Glucoside in cultivated and wild Gentiana officinalis H.Smith can reducethe phosphorylation of P38, ERK, JNK in diffrernt degrees, Rat chondrocytesstimulated by IL-1β showed increased PGE2, COX-2, MMP-1, MMP-3and MMP-13levels, whereas the expression of Collagen type II decreased.However, this activationwas inhibited by co-incubation with various concentrations of Gentiopicroside.Analysis by HPLC-UV/MS, combined with retention time of standard substances,literature and mass spectrometric data, wild Gentiana officinalis H.Smith affordedeight iridoids, identified as loganic acid, qinjioside A, shanzhiside methyl ester,6’-O-β-D-glucosyl gentiopicroside, swertiamarin, gentiopicroside, sweroside,dehydropentstemoside. Cultivated Gentiana officinalis H.Smith afforded seveniridoids, identified as sweroside, sesamoside, loganic acid, shanzhiside methyl ester,6’-O-β-D-glucosyl gentiopicroside, swertiamarin, gentiopicroside.The study of total secoiridoid glucoside and gentiopicroside on chondrocytesinduced by IL-1β in vitro, discovered that its reduced the phosphorylation of P38,ERK, JNK in diffrernt degrees, the eapression of COX-2and the production of PGE2,down-regulated the expression of MMP-1, MMP-3, MMP-13.Through the process tosuppress collagen type Ⅱ degradation and finally to protect chondrocytes. Further,analysis by HPLC-UV/MS,the effective substances were loganic acid,qinjioside A,shanzhiside methyl ester,6’-O-β-D-glucosyl gentiopicroside, swertiamarin,gentiopicroside, sweroside, sesamoside.
