Coleus forskohlii (Willd) Briq.(Labiatae), which mainly distriubtes in Indian andYunnan Province of China, is a folk medcine against asthma, cough, bronchitis, etc.It’s used mainly by Chinese people to treat cough, dizziness and other symptoms. Inthis thesis, we report the separation and structure identification of C. forskohlii inchemical composition, as well as initial studies with anti-hepatic injury activity of itscrude extract.The chemical constituents were isolated and purified by Silica gel, RP-C18andSephadex LH-20column chromatography, as well as the semi-preparative RP-HPLC.The structures were determined on the basis of NMS and MS analysis. A total of40compounds were isolated from C. forskohlii. However, only21compounds wereelucidated. They are Forskoditerpene F(CF-14), Apigenin(CF-09),Stigmasterol-3-O-β-D-glucopyranoside (CF-23), Lupeol(CF-25),19α-hydroxyerythrodiol(CF-28), β-Sitosterol(CF-01), Isoforskolin(CF-02), Forskolin H(CF-03),Forskolin A(CF-04)、14-deoxycoleon U(CF-05), Forskolin I(CF-07), ForskolinJ(CF-10), Forskolin E(CF-11),12-hydroxy-8,13E-labdadien-15-oic acid(CF-12),Betulinic acid(CF-13), Coleolic acid(CF-15), Coleonic acid(CF-22),6-acetyl-1,9-dideoxy-forskolin(CF-24), Euscaphic acid(CF-27), Forskolin G(CF-29),Forskolin K(CF-34).A initial study on the C. forskohlii biological activity of different solvent extractwas carried out. Using CC14induced acute liver injury as model in mice. Study C.forskohlii hepatoprotective activity of ethyl acetate extract, n-butanol extract andwater extract. The hepatoprotective activity was assessed using various biochemicalparameters, such as aspartate aminotransferase (AST), alanine aminotransferase(ALT), alkaline phosphatase (ALP), direct bilirubin (DB), unconjugated bilirubin(UB), total bilirubin (T-Bili) vitality, and histological examination of livers wascarried out. Those results showed that the n-butanol extract repair liver damage better,significantly reduced serum AST, ALT, A LP, DB, UB, TBIL activity (P<0.01);Pathological changes in hepatic tissue were observe under an optical microscope, the mouse model of liver injury was significantly changed: loss of normal structure, partof the liver lobule liver cells have different degrees of necrosis with inflammatory cellinfiltration, partially visible increase in size, was vacuoles flaky lesions, comparedwith the normal group there are significant differences, is successful modeling.Silymarin group shown to have positive effects in treating liver injury: no necrosis.N-butanol extract high dose group showed a significant decrease in hepatic lobulesincomplete, irregular arrangement of hepatic cords, necrosis and inflammatory cellinfiltration, From this sequence of events it is obvious that liver regeneration.Meanwhile, it may be inferred that C. forskohlii extract on the liver cells have aprotective effect.
