| To investigate the role of miR-126in the pathogenesis of preeclampsia by studyingthe different expression of miR-126in endothelial progenitor cells from cord blood between patients with preeclampsia and normal pregnancy, and the effects of different expression of miR-126on the angiogenic ability of EPCs. Methods1. We chose twelve normal pregnant women and twelve patients withpreeclampsia,and collected the basic clinical data.2. Endothelial progenitor cells were separated and cultured for seven days, and thenidentified by cell morphology and immunofluorescence.3. Real-time quantitative PCR was used to detect the expression of miR-126.4. Plasmid with miR-126mimics or with mimics control was transfected intoendothelial progenitor cells as experimental group,negative control group,and the blank control group had no transfection. 5. Real-time quantitative PCR was used to detect the expression of miR-126in transfected EPCs.6. Change of differentiation was detected by observing the counts of spindle cells.7. Change of migration was detected by transwell test.8. Change of tube formation capacity was detected by tube formation.Results1. Expression of miR-126in preeclampsia was decreased by (64.3±9.5)%,which was significantly different(P<0.05).2. Expression of miR-126of transfected EPCs was increased by (735.50±40.35)%,which has a statistically difference(P<0.05).3. The amount of differentiated cells in experimental group, negative control group and blank control group were130±3.5,59±2.6and53±3.0,respectively. The experimental and negative control had a statistically difference (P<0.05),while the negative and blank control group had no obvious difference (P>0.05).4. After EPCs were seeded24hours, the amount of migration cells in experimental group, negative control group and blank control group were128.4±7.5,70.6±4.7and83.3±4.2,respectively.The experimental and negative group had a statistically significant difference (P<0.05),while the negative and blank control group had no obvious difference (P>0.05).5. After EPCs were seeded24hours, the amount of branch points in experimental group, negative control group and blank control group were59±6.7,26±3.5and29±4.3, respectively. The experimental group and the negative group had a statistically significant difference (P<0.05),while the negative and blank control group had no obvious difference (P>0.05).ConclusionThe study shows that the expression of miR-126is decreased in EPCs of patients with preeclampsia and over-expression of miR-126can improve the proliferation,migration and tube formation capacity of EPCs. So we inference that the decrease of miR-126in EPCs may be one of the possible causes for the pathogenesis of preeclampsia. |
