Multifunctional NPs PTX-PLA-QDs-MNPs were prepared by hand-driven membrane-extrusion emulsification approach, and we studied the effect of the above NPs on human malignant melanoma A875cells including cellular uptake, cytotoxicity, and magnetic resonance imaging (MRI) studies.Methods:1. Uniform biodegradable polymer nanoparticles (NPs) with controllable sizes were prepared by hand-driven membrane-extrusion emulsification approach. Morphology of nanoparticles was characterized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Size and size distribution (PDI) of the NPs were analyzed by dynamic light scattering (DLS). The encapsulation efficiency (EE%) of NPs was measured by HPLC and TEM. Drug release profile of the NPs in vitro was investigated by HPLC and dialysis.2. Further experiments of NPs in vitro A875cells were carried out, including cellular uptake by fluorescence microscope, cytotoxicity by CCK-8assay, and magnetic resonance imaging (MRI) studies by MRI scans. This approach could be extended to the preparation of PCL and PLGA NPs.Results:1Uniform biodegradable polymer nanoparticles (NPs) were easily generated by this simple method-hand-driven membrane-extrusion emulsification approach;2. In contrast to PBS and PLA-NPs, after incubation with PTX-PLA-QDs-MNPs for10min, weak red fluorescence signal was observed in the cytoplasm and around nuclei, indicating the appearance of NPs in the cells; after6h incubation, we observed strong fluorescence in the cytoplasm and red fluorescence spot in the nucleus;3. Compared with native PTX and PTX-PLA-QDs, PTX-PLA-QDs-MNPs displayed more obvious cytotoxicity, while PTX-PLA-QDs showed stronger cytotoxicity than PTX;4. With the increase of iron concentration, the signal intensity of MRI decreased accordingly. Silimar results could also be obtained for the preparation of PCL and PLGA NPs.Conclusion:PTX-loaded NPs prepared by this simple method are multifunctional, and promising in drug loading, bioimaging, and tumour therapy (e.g., MM therapy).
