Objective To investigate the genes related to the process of renal carcinoma cell OS-RC-2bone metastasis,so as to provide us with new approach to clinical therapy..Methods Establish the animal model of renal carcinoma cell OS-RC-2bone metastasisby injecting OS-RC-2into the right tibia of nude mice anesthetized with i.p. sodiumpentobarbital and gain tumor cells from metastasis lesions,comparing with OS-RC-2in abilityof proliferation and invasion by MTT and Transwell. Screening technology was used todetect differentially expressed genes between the original tumor and metastasis.Investigaterelated genes with KEGG databases,RT-PCR and Western blot are performed subsequentlyto verify the result of microarray analysis.Results Build the animal model of renal carcinoma cell OS-RC-2bone metastasis andharvest the bone seeking renal carcinoma cell OS-RC-2-BM-3.The data of microarrayanalysis revealed a signifcant upregulation of926genes and downregulation of270genesin the OS-RC-2-BM-3clone compared with the OS-RC-2cell line.HIF-1α and HSP90were screened out through KEGG databases, the results of RT-PCR and Western blotcorrespond to the data from microarray analysis.Conclusion The process of renal carcinoma cell OS-RC-2bone metastasis isassociated with multiple genes,and HIF-1α and HSP90may play a crucial role in thisprocess.
