Objective: To construct targeting prostate cancer gold nanocapsules(GNCs) loading99mTcand evaluate i ts binding affinity and cytotoxicity toward human prostate Cancer linePC-3were assessed in vitroMethods: the formation of gold hollow nanocapsules(GHNCs)is attributed to twosubsequent reduction reactions: the initial reduction of HAuCl4by Co nanoparticles,followed by the reduction by NaBH4,and enwraped the radionuclide99mTc in the GHNCsto form the99mTc-GHNCs,finally,the99mTc-GHNCs reaction with the FA-PEG-SH tofabricate the FA-PEG-GHNCs-99mTc. We characterize the FA-PEG-GHNCs-99mTc bytransmission electron microscope (TEM), emission scanning electron microscope (SEM)respectively. Radioligand binding assay of FA-PEG-GHNCs-99mTc were explored byincubated PC-3cells with medium contained different concentration (0.1nM,1nM,10nM,20nM,40nM,60nM,80nM,100nM) of FA-PEG-GHNCs-99mTc for2h, the boundradioactivity was measured in a γ-counter. For the cytotoxicity reaserch, PC-3cells werecultured with medium contained different concentration (1nM,10nM,20nM,40nM,60nM,80nM,100nM) of FA-PEG-GHNCs-99mTc for24h, the cytotoxicity effect was tested byMTT method. Apoptosis was assessed by flow cytometry, PC-3cells were exposed todifferent concentration(20nM,40nM,80nM) of FA-PEG-GHNCs-99mTc in culture mediumfor24h, trypsinized, centrifuged, re-suspended, and then add5μl Annexin V-FITC and5μlPropidium Iodide, Samples were analyzed using a flow cytometer. Resμlts:1.Binding parameters (Kd=39.42and Bmax=2202) were calculated usingGraphPad PRISM5.0.2.The Relative growth rate (RGR)was105.174%,103.487%,105.999%,99.587%,90.251%,84.217%,80.278%,respectively in different concentrationof FA-PEG-GHNCs-99mTc groups,and the CytoxicityGrade was0,0,0,0,Ⅰ,Ⅰ,Ⅰ.3. theapoptosis rate of PC-3cells was8.97%±1.71%,9.71%±3.50%,10.37%±3.64%,respectively in different concentration of FA-PEG-GHNCs-99mTc groups.Conclusion: The FA-PEG-GHNCs-99mTc targeting folate receptors of bind to PC-3cells invitro, No significant cytotoxicity was observed in the PC-3cells at the low concentrationsof FA-PEG-GHNSs-99mTc, And no significant increase of cell apoptosis was detected in thePC-3cells treated with FA-PEG-GHNCs-99mTc, which indicates good biocompatibility andgreat potential in tumor-targeted drug delivery and nuclear imaging. It laid the foundationfor multifunctional gold nanocapsule mediated treatment of prostate cancer underSPECT/CT guidance.
