Objective: By studying the effects of Angelica flavescens Fritillaria pillmedicated serum on gastric cancer SGC-7901, MKN-45proliferation and invasionand metastasis, it is to investigate the effects of different concentrations of Angelicaflavescens Fritillaria pill medicated serum on proliferation, cell cycle, invasion andmetastasis of gastric cancer cell lines SGC-7901and MKN-45with its mechanismsinvovled.Methods:After treated with different concentrations of Angelica flavescensFritillaria pill medicated serum, gastric cancer cell lines SGC-7901and MKN-45weretested by the following methods:(1) MTT was employed to test the vitality of gastriccancer cell lines SGC-7901and MKN-45;(2) flow cytometry was used to detect cellcycle;(3) Tran-swell and scratch adhesion test was used to detect the invasion andmetastasis abilities;(4) qRT-PCR technique was used to detect the expression ofCOX-2, BCL-2, BAX and PTEN gene;(5) Immunocytochemistry was employed totest the expression levels of protein COX-2, BAX and PTEN.Results:(1)Angelica flavescens Fritillaria pill medicated serum of differentconcentrations can inhibit the proliferation of SGC-7901and MKN-45cell lines, andmedicated serum of10%concentration reached a maximum inhibition, when treatingon SGC-7901cell for72h, the inhibition rate of high-dose group, middle dose groupand low dose group were34.51%,31.69%and19.78%respectively; when treating onMKN-45cell for72h, the inhibition rate of high-dose group, middle dose group andlow dose group were35.66%,34.27%and22.38%respectively, the differenceshowed statically significant (P <0.05);(2)Flow cytometry analysis showed: when treated by high-dose of Angelicaflavescens Fritillaria pill medicated serum for72h, SGC-7901cell in S phaseincreased from46.5%down to13.7%, G1/G2phase increased from53.5%to74.1%rise; MKN-45cell in S phase from44.9%down to9.94%, G1/G2phase ratio rosefrom55.1to72.1%; (3)Transwell and scratch adhesion test showed: when treated on SGC-7901cellby high-dose for72h, the ability of invasion decreased40.48%, the ability ofmetastasis and migration decreased46.88%and41.30%respectively; when treated onMKN-45cell by high-dose for72h, the ability of invasion decreased37.23%, theability of metastasis and migration decreased50%and36.46%respectively; thedifference showed statically significant (P <0.05);(4)qRT-PCR results showed: when treated on SGC-7901cell by high-dose for72h, the expressions of COX-2and BCL-2mRNA decreased68%and77%; and theexpressions of PTEN mRNA and BAX mRNA increased99.7%and85.6%; whentreated on MKN-45cell by high-dose for72h, the expressions of COX-2and BCL-2mRNA in decreased90%and91%in SGC-7901cell respectively, and the expressionsof BAX and PTEN mRNA expression levels increased98.65%and89.72%, thedifference showed statically significant (P <0.05);(5)Immunocytochemistry experiment showed:containing serum can increasedthe average gray value of COX-2of SGC-7901cells, and reduce its positive rate,reduce the average gray value of BAX and PTEN, and increased its positiveexpression rate; containing serum can increased the average gray value ofCOX-2of MKN-45cells and and reduce its positive rate, reduce the average grayvalue of BAX and PTEN of BAX and positive PTEN, and increased its positiveexpression rate, the difference showed statically significant (P <0.05, except blankserum).Conclusion: Angelica flavescens Fritillaria pill medicated seruP <0.05m couldinhibit the proliferation of SGC-7901and MKN-45cell lines, weakening its invasionand migration capacity and the mechanisms of which may be related to theintervention of cell cycle, the genes and proteins of COX-2, BAX, BCL-2and PTEN.
