Objective:To observe the neuroprotective effects of isoliquiritigenin (ISL) on IL-6, TNF-a,SOD, and MDA in the brain tissue and neurobehavioral after traumatic brain injury (TBI) in rats,and to explore the underlying mechanisms of ISL treatment on the TBI.Methods: Rats were randomly divided into3groups: A group (sham-operated group), Bgroup (model group) and C group (ISL-treated group);each group have15rats, total of45ratsfrom60rats were screened. TBI model was established with the modified Feeney’s method. Thetreatment group received isoliquiritigenin1%Tween8030mg/kg intraperitoneal injection of asaline suspension1times/day, the sham group and model group were given the same volume of1%Tween80saline by intraperitoneal injection of1times/day of treatment5days. After5d TBI,the rats were sacrificed to measure the brain water content with wet-dry weight method. Thecontents of MDA, and the activity of SOD in the brain tissue were detected with biochemistrymethods. The contents of IL-6and TNFa in the brain tissue were detected with ELISA methods.The morphological change of neuron in hippocampal area and cortex was observed under opticalmicroscope through H-E staining. The neuron behavioral observation by balance beam walking testand bilateral forepaws grasps test.Result: Compared with B group (model group), the water content of brain was decreased, thedamage of hippocampal area was alleviated, the activity of SOD was elevated, and the content ofMDA, IL-6and TNF-a were reduced, the balance beam walking test score were lowered, the frontpaws strength score were increased, Neurological was improved, significantly (P <0.05) in Cgroup (ISL-treated group).Conclusion: ISL can increase the activity of SOD, reduce MAD generation, reducing TBIbrain tissue oxidative stress, inhibition of inflammatory cytokines IL-6and TNF-a production andrelease, reducing inflammation on brain tissue of TBI, improve limb muscle strength and motorcoordination on TBI rat. ISL can reduce the water content of brain in TBI rat, to promote traumaticlesions recovery, protection of nerve tissue, improve neurological behavior. The neuroprotectiveeffects of ISL on TBI rats may be by inhibiting oxidative stress and inflammatory cytokineproduction and release.Signification: ISL has significantly neuroprotective effects in the experiments on rats with TBI, so this experiment provides a theoretical basis for ISL developed as a neuroprotective drugsin clinical TBI that for clinicians treating TBI provides a new idea.
