Objective:(BMSCs) cell sheet with bone marrow mesenchymal stem cells wrapped intosilk fibroin (Silk fibroin, SF)/chitosan (chitosan, CS)/BMSCs after induction of complexthree-dimensional scaffold formed to observe and detect and discuss osteogenic effect ofthe complex.Methods: Density gradient centrifugation and cultured rabbit bone marrow mesenchymalstem cells to differentiate into bone cells induced differentiation direction, temperaturereactive inoculated petri dishes,37℃,5%saturated humidity training, and then cooled to20℃preparation of cell sheets layers. Will be prepared after BMSCs cell sheet withSF-CS/BMSCs induced biological support structure built complex. The experiment wasdivided into group A, B, A group after BMSCs cell sheet/SF-CS/BMSCs inducedbiological support structure built complex, group B after SF-CS/BMSCs inducedbiological stent placed in the2groups Petri dishes at37℃,5%saturated humidity vitrodrawn on days3,8,14, quantitative alkaline phosphatase activity, scanning electronmicroscopy and other testing.Results: Successfully constructed BMSCs cell sheet, after BMSCs cell sheet/SF-CS/induced BMSCs scaffold complexes. A group than in the group of alkaline phosphatasequantitative B, ELLSA determination of osteocalcin in group A than in group B, there aresignificant differences (P <0.05), scanning electron microscope examination showedmatrix mineralization nodules A group of more than group B.Conclusion: BMSCs cell sheet and SF-CS/BMSCs induced composite scaffoldosteogenesis than SF-CS/scaffold induced BMSCs, complexes such expected becomebetter bone tissue engineering.
