Objective: Apply DNA barcode to identify Miao medicine Blumea balsamifera, itsclosely related species and adulterants, to determine the validity of identification inChinese herbs.Methods: Total genomic DNA was isolated from B.blasamifera and adulterants. NuclearDNA ITS, psbA-trnH, matK, rbcL sequences were amplified and purified PCR productswere sequenced. Sequences assembly and consensus sequence generation were performedusing the CodonCode Aligner V3.0. The Kimura2-Parameter (K2P) distances werecalculated using the MEGA5.0. Identification analyses were performed with usingBLAST1, nearest distance (ND) and neighbor-joining (NJ) methods.Results: The ITS and psbA-trnH complete sequence, rbcL and matK partial sequence ofB.blasamifera were got. K2P genetic distance between species within is much larger thanthe smallest species of the largest K2P genetic distance ITS, ITS2, psbA-trnH threesequences from closely related species and their adulterants. NJ tree was constructed basedon the three sequences, B.blasamifera their relatives were clustered together, and they weremonophyletic, obviously with adulterants Callicarpa macrophylla.Conclusion: Compared with the rbcL, matK, psbA-trnH sequence, ITS2sequences issuitable for identification of Miao medicine B.blasamifera its closely related species, andfor identification of B.blasamifera adulterants. The method provides a scientific basis.
