Objective: DWI, in combination with bone metabolize and histopathological were usedto study the ONFH in the pre-early stage on a model of rabbit which was injected withhormone and to explore the diagnostic value of DWI in the pre-early diagnosis of ONFH.Methods:40healthy rabbits were randomly divided into two groups,20for control groupand the remaining20for experiment group. The experiment group was injected horseserum10ml/kg through the ear vein at the first time and after2weeks injected5ml/kg oncea day for2consecutive days. When injected the horse serum at the first time,and theninjected methylprednisolone4mg/kg,Latter24hours,once a week for8consecutive weeks.The control group were injected NS with the same methods at the same time. At the2、4、6、8weekends, both the control group and the experiment group were inspected withfollowing ways: First of all, drawing off the blood to assay TRACP-5b、BGP, and then todo MRI and DWI. At last, removing the femoral head to do the histopathologicalexamination.Results: Bone metaplasia findings: compared with the control group: the TRACP-5b ofthe experiment group was obviously increased and the BGP was obviously decreased in the4,6,8week (P<0.05),the experiment group’s TRACP-5b and BGP didn’t have any changein the2week(P>0.05); Compared with the experiment group in the2week: theTRACP-5b of the experiment group was obviously increased and the BGP was obviouslydecreased in the4,6,8week (P<0.05); Compared with the experiment group in the4week: the TRACP-5b of the experiment group was obviously increased and the BGP wasobviously decreased in the8week (P<0.05), the TRACP-5b and the BGP didn’t have anychange in the6week (P>0.05); Compared with the experiment group in the6week: theTRACP-5b of the experiment group was obviously increased and the BGP was obviouslydecreased in the8week (P<0.05); TRACP-5b and BGP of the control group’s didn’t have any change in each time. MRI findings: MRI and DWI for control group and experimentgroup did not show any abnormality during each time; ADC was obviously higher thancontrol group;As time went on, the ADC was gradually raised(P<0.05). Pathologicalfindings: at the secend week nucleus poly edge was appeared, and the number of emptylacunae was increasing when compared with the control group. AT the forth week, bonetrabeculae were shriving、 thining and derangement.The number of osteoblasts wasobviously reducing and the number of empty lacunae was obviously increasing, fat cellswere bigger than before and some of them were fused. From the sixth to eighth week, bonetrabeculae reduced、thickened and fractured, bone nucleus were disappeared, and therewere a large number of empty lacunaes. There were a little of osteoblasts near the bonetrabeculae, marrow cavity was occupied with fat cells and some of them were fused.Conclusion:1、The model that was made by Horse serum and some hormone can be usedto research the ONFH in the pre-early stage;2、The pecific indicators of TRACP-5b andBGP has references value to diagnosis the ONFH in the pre-early stage;3、The increase ofADC can reflect the occurrence and progress of the ONFH in the pre-early stage, whichcan provide valuable information for clinical diagnosis.
