Objective: To explore suitable culture condition of type I and type II muscle cellthrough the expression of MHC isoforms of cell cultured in vitro.Methods:The inherent semimembranosus compartment and sub-semimembranous dorslecompartment were abtained from eighteen rabbit,4-6weeks old. These samples weredivided4groups as: type I and II skeletal muscle tissue group, type I and II skeletal muscledigestive cell group, type I and II skeletal muscle cell normal culture group, type I and IIskeletal muscle hypoxic(3%O2) culture group. The normal culture group and hypoxicgroup were cultured with high glucose DMEM containing15%fetal calf serum. MHCisoforms were respectively detected at1、3、7day by RT-PCR.after cell cultured.Results:1. Inherent semimembranosus express type I and a few of MHC IIa, Sub-semimembranosus tissue express MHC IIa, MHC IIb.2. Non-cultured inherent semimembranosus cells express MHC I、MHCIIa and MHC IIb,non-cultured sub-semimembranosus cells express MHC IIa, MHC IIb.3. Inherent semimembranosus cell normal culture group express MHCⅡa, MHCⅡb type,not MHC I, and sub-semimembranosus express MHC IIa, MHC IIb type, not MHC I.Under condition of hypoxia, two different group of skeletal muscle cells express MHCⅡa,MHCⅡb or MHCⅡx type, not MHC I.4. Statistical analysis shows:compared with muscle tissue, MHC I expression of Type ISkeletal muscle cell in normal culture group and in hypoxic culture group all exist obviousdifference (P<0.05), while expression of MHCⅡa、MHCⅡb and MHCⅡx not obviousdifference. compared with muscle tissue, expression of MHCⅠ、MHCⅡa、MHCⅡband MHCⅡx exist no obvious difference in Type II Skeletal muscle cell of normal cultureand hypoxic culture (P>0.05). Conclusions:1.High glucose DMEM containing15%fetal calf serum is suitable forskeletal muscle cell growth;2. Two different group of skeletal muscle cell had a trend of translating type I fibre intotype II fibre.3.Hypoxic culture condition promote the translation of type I fibre into type II fibre.
