Capillaris is a traditional medicinal and edible plant in China. The main effect of capillaries is protecting liver and choleretic. Both Artemisia capillaris Tunb. and Artemisia scoparia Waldst. et Kit. can be treated as capillaries. There are lots of reports are about small molecule substance of capillaries, but the study on polysaccharide of capillaries are seldom reported.The capillaris in this study is Artemisia capillaris Tunb. which collected in Ningshan (a county of Shannxi). We took modern techniques to extract, isolate and purify the polysaccharides from Artemisia capillaris Tunb. which collected in different harvest time, and studied the homogeneous compositions’ structures. We also compared the antibacterial activities of the two capillaris polysaccharides. The main conclusions are as follows:(1) Extracting polysaccharides from Artemisia capillaris Tunb. by hot water extraction, and handling them with decoloration, deproteinization and degreasing, we got polysaccharide from capillaris which collected in spring115g and polysaccharide from capillaris which collected in autumn115g. Both of them contain no proteins and nucleic acids.(2) The results of antibacterial activities experiments show that both of the two capillaris polysaccharides have inhibitions on normal pathogenic bacterial in food. The antibacterial activity of autumn capillaris polysaccharide is better than the spring capillaris polysaccharide. The inhibitions on bacteria of the two capillaris polysaccharides are better than fungi. The antibacterial activities of the two capillaris polysaccharides will be decreased or have no inhibitions when pH value less than6or greater than7. (3) By DEAE-52cellulose column chromatography and Sephadex G-100column chromatography, one composition named KY-1is isolated and purified from spring capillaris polysaccharide, and two compositions named KY-2, KY-3are isolated and purified from autumn capillaris polysaccharide. The HPLC proved that all the three compositions are homogeneous components.(4) Determined by HPLC, the molecular weights of KY-1, KY-2and KY-3are5.23×104Da、1.93×104Da、2.28×105Da.(5) Paper chromatography shows that KY-lis composed of arabinose, glucose and xylose; KY-2is composed of glucose, mannose and rhamnose; KY-3is composed of glucose, mannose and xylose.(6) IR and’HNMR show that KY-1, KY-2and KY-3contain hydroxy (3400cm"1) and α-pyran ring anomeric hydrogen(1000cm-1,δ>5.00ppm).(7) The physical and chemical properties experiment shows that all the three compositions are acidic polysaccharide. They have similar physical and chemical properties and optical activity.
