| Background In recent years, with the depth research on themolecular biology,effect of various growth factors in the fracture healinghas become one of the hot,such as BMP-7ã€TGF-β1and bFGF. DANSHENis a representative of the Chinese medicine drugs for improving bloodcirculation,many scholars have made an in-depth study on the treatment offractures using DANSHEN,and achieved encouraging results.However,wedid not know the changes of BMP-7TGF-β1bFGF in the healing processof rabbits fracture using the “DanShen†injection.Objective To determine the changes of BMP-7ã€TGF-β1and bFGFin the healing process of rabbits’ fracture after using “DanShen†injection,and explore the molecular biology mechanism of promoting fracturehealing of “DanShen†injection.Methods Divided72rabbits of New Zealand into experimentalgroup and control group randomly.All of the rabbits were made intofracture model.“DanShen†injection was injected into the experimentalgroup,normal saline was injected into the control group.Extracted serum and accepted X-ray examination to evaluate the the healing of fracture at1,2,3,4,6,8week postoperatively.ELISA was used to determine the serumcontent of BMP-7ã€TGF-β1and bFGF.Callus of fracture site were deal withHE staining to observe the healing of fracture and immunohistochmicalmethod was used to observed the expression of BMP-7ã€TGF-β1and bFGF.All data was analyzed by SPSS18.0statistical software and there wassignificant differenc when P<0.05.Results X-ray shows experimental group’s bone callus appearedearlier and having more quantity bone callus at3,4weekpostoperatively.The experimental group’s fibronectin andosteoblast occursearlier than control group through HE at3,4week postoperatively.The twogroups serum content of BMP-7ã€TGF-β1and bFGF had significantdifferenc at4,2,3week respectively and immunohistochmical stainingdisplayed that the content of BMP-7ã€TGF-β1and bFGF had significantdifferenc at3,2,3week respectively.Conclusion “DanShen†injection can promote expression of theBMP-7ã€TGF-β1and bFGF during healing of fracture. |
