ObjectiveTo study the effect of MLN4924on proliferation and apoptosis of lungepithelial carcinoma A549cells, and the effect of MLN4924combined withDDP on the A549cells.MethodsA549cells were treated with MLN4924and DDP at differentconcentrations and incubation time. The inhibition rate of cell growthdetected by cell counting kit-8(CCK-8) assay, the apoptosis by Hoechst33342and flow cytometry, and the protein expression of cell apoptosisassociated proteins Caspase-3, NF-κB p65and CDT1by Western blot.ResultsThe proliferation of A549cells was decreased while increasing ofconcentration or prolonging the incubation time. MLN4924induce S phasecell cycle arrest. The protein expression of cell apoptosis associatedproteins Caspase-3and CDT1is higher than the control group. NF-κB p65is lower than the control group. ConclusionMLN4924effectively inhibited proliferation and induce apoptosis inA549cells. The relevant mechanisms induce S phase cell cycle arrest,increased caspase-3protein expression, and inhibition of NF-κB pathwayactivation.MLN4924and DDP have synergistic effect on lung cancer lineA549proliferation inhibition.
