Objective: To evaluate the oxidative status of cases including NGT,IGR and T2DM, and study the relationship between oxidative stress derivedfrom Nox and pancreatic β-cell function.Methods:84citizens came for routine physical examination in ourhospital were selected. The FPG, FINS, HbA1c, serum lipids, Nox,8-OHdG,SOD, MDA, blood pressure, height, weight, waist circumference and hipcircumference were detected or measured of each case. The OGTT andOGIRT were completed according to the standard operation procedures, theplasma glucose and insulin were detected at0min,30min,1h and2hpoints.The HOMA-β, HOMA-IR, HBCI, BMI and WHR were calculated byeach recognized formula.Results: The84research objects were divided into3groups accordingto the OGTT results: NGT group, IGR group and T2DM group.(1)The levelof Nox,8-OHdG and MDA lifted from NGT to T2DM, while the level ofSOD reduced in turn, the differences were statistically significant (P<0.01).(2)The degree of HOMA-IR increased in turn, but HOMA-β showed a opposite manifestation, both of the two had statistically significantdifferences (P<0.01). And the difference of HOMA-β were much morepronounced after corrected by HOMA-IR, VIZ namely HBCI (P<0.01).(3)The linear correlation analysis (Nox as the dependent variable) showedthat there’s a significant positive correlation between Nox and WHR, HbA1c,TG, SBP,8-OHdG, MDA(P<0.05~P<0.01). There’s a significant negativecorrelation between Nox and HDL-L, HBCI, SOD(P<0.05~P<0.01). Thelinear correlation analysis (HBCI as the dependent variable) showed thatthere’s a significant positive correlation between HBCI and SOD(P<0.01),there’s a significant negative correlation between HBCI and BMI, WHR,HbA1c, SBP, Nox,8-OHdG, MDA(P<0.05~P<0.01). The further multiplestepwise linear regression analysis (HBCI as the dependent variable)indicated that only Nox had a significant effect on HBCI, the rests didn’tenter into the equation of multiple stepwise linear regression.Conclusions:(1)With the ascent of body blood glucose concentration,the Nox were over activated, the level of oxidative stress elevated, oxidativestress injury aggravated and body antioxidant capacity decreased.(2)Compared with other traditional indexes of oxidative stress, Nox is muchmore ideal in reflecting the oxidative status in the body.(3)Oxidative stressderived from Nox is closely tied to pancreatic β-cell function, but theconcrete mechanism of the interaction between them remain unknown.Probably it would be a effective way to improve pancreatic β-cell function and treat type2diabetes mellitus by regulating the oxidative status with thetarget of Nox.
