The Relationship Between Antinuclear Antibody And Autoimmune Liver Disease Antibody With Hepatitis B

Objective: To research the relationship between autoantibodies andthe progress of hepatitis B. Methods:The antinuclear (ANA)，nRNP/sm，sm，dsDNA，ribosomal protein P，SSA，SSB，RO52and autoimmune hepatitisassociated autoantibodies such as anti smooth muscle antibody(ASMA),liverkidney microsomal antibody(LKM),antimitochondrial antibodies M2(AMA-M2)were detected with indirect immunofluorescence and immunoblotting.HBV-DNA and HBV gene were tested by quantitative PCR typing.Automaticbiochemical and chemiluminescence analyzer were useted to detect serumALT,AST and the index of liver fibrosis.And then the cytokin ofIL-5,IL-4,IL-10were detected with ELISA.Resμlts:①nRNP/sm, SSA,SSB,RO52, SCL-70were12.6%,6.3%,5.3%,9.5%,7.4%in CHB; nRNP/sm, SSA,SCL-70were1.5%,1.5%,1.5%in control group.The ANA positive rate of CHBwas23.2%, significantly higher than4.5%of the control group,mainly in lowtiter and nuclear particles of karyotype.②The ASMA,LKM and AMA-M2antibodies were finded in CHB group and the LC,SLA were suspiciouspositive; however， no such antibodies in control group.③ALT,AST,viral loadand HBV genotypes were116.2IU/L,86.3IU/L,4.8,21/25(HBV B),3/25(HBV C)in CHB with antibody positive group; ALT,AST,viral load and HBV genotypeswere108.3IU/L,91.3IU/L,4.9,62/70(HBV B),9/70(HBV C) and1/70(HBV other type)in CHB with antibody negative group. Two groups of resμlts was notstatistically significant (P>0.05).④the serum type III collagen,the serum typeIVcollagen,aminin LN,hyaluronic acid enzyme HA,IL-5,IL-4,IL-10were191.2ug/l,116.7ng/ml,131.5ng/ml,1125ng/ml,4.9ng/ml,6.2ng/ml,14.3ng/ml inCHB with antibody positive group; the serum type III collagen,the serum typeIVcollagen,aminin LN,hyaluronic acid enzyme HA,IL-5,IL-4,IL-10were172.3ug/l,65.8ng/ml,87.2ng/ml,726.3ng/ml,4.2ng/ml,6.4ng/ml,11.2ng/ml inCHB with antibody negative group.Compared with CHB of antibodynegetive,the serum type IVcollagen,aminin LN,hyaluronic acid enzyme HA andIL-5,IL-10in CHB with antibody positive group were higher(P<0.05).But theserum type III collagen and IL-4had no significant difference in twogroup(P>0.05). Conclusion:our resμlts suggest that the autoantibodies are notonly confined to the antinuclear antibody, but also autoimmune hepatitisassociated autoantibodies in hepatitis B, the presence of autoantibodies maycause liver cirrhosis induced by hepatitis B.
