Objective:To investigate the expression of human epidermal growth factor2(HER2) intissues of colon cancer.To study the correlation of HER2expression level andchinicopathological characteristics of colon cancer.Methods:The experimental group consisted of108cases of colon cancer tissues.Meanwhile,108cases of normal tissues taken from the corresponding area around thecancer were treated as the control group. The Real-time quantitative PCR (RTQ-PCR)and immunohistochemistry (IHC) were used to detect the expression of HER2incarcinomatous tissues and adjacent normal tissues.Results:RTQ-PCR results showed that: HER2mRNA in colon cancer tissues was higherthan that in adjacent normal tissues. HER2mRNA in colon cancers of clinical stageIII/IV was higher than that in the clinical stageⅠ/Ⅱ group. HER2mRNA in thecolon cancer group with lymph node metastasis was higher than that without lymphnode metastasis. The difference was statistically significant (P<0.05). IHC resultsshowed that:108cases of colon cancer tissues, HER2-positive expression in24cases.No case of adjacent normal tissue have HER2-positive expression, the difference wasstatistically significant (P<0.05). HER2-positive rate in lymph node metastasis group(36.8%) and clinical stage III/IV group(26.5%) was higher than that in no lymph nodemetastasis group(14.3%) and clinical stage I/II group (15.0%). The difference wasstatistically significant (P<0.05).But HER2-positive rate was not correlated withgender, age, tumor size and tumor differentiation.Conclusion:HER2was over-expressed in colon cancer. HER2expression levels was closelycorrelated with clinical stage and lymph node metastasis.
