Objective:To investigate the expression and significance of Von Hippel–Lindau(VHL)and vascular endothelial growth factor(VEGF) in human renal clear cellcarcinoma(RCCC),and the relationship between their expression.Methods:Formalin-fixed paraffin embedded tissues from45cases of RCCC and19casesof normal renal tissues which were closely adjacent to renal clear cell carcinoma and11cases of normal renaltissues were evaluated for Von Hippel–Lindau proteinexpression by immunohistochemical S-P method.The results were compared withhistological grade and clinical stage. vascular endothelial growth factor was alsodetected in45cases of RCCC and19cases of normal renal tissues which wereclosely adjacent to renal clear cell carcinoma and11cases of normal renaltissues andits correlation with VHLwas concerned.Results:The positive incidence of expression VHL protein in RCCC was44.4%(20/45)which was prominently lower than those in the adjacent tissues of RCCC[89.5%(17/19)] and normal renal tissues [100%(11/11)](P<0.05).The expression ofVHL was negatively correlated with histological grade clinical stage andprognosis.The positive incidence of expression VEGF protein in RCCC was73.3%(33/45)which was prominently higher than those in the adjacent tissues ofRCCC[26.3%(5/19)] and normal renal tissues [18.2%(2/11)](P<0.05).Theexpression of VEGF was notable correlated with histological grade and clinicalstage.there is trend that VHL negatively correlated with VEGF(p<0.05).Conclusion:Von Hippel–Lindau (VHL) of the abnormal protein in renal clear cellcarcinoma clinical stage, pathological grade, infiltration plays a key role in thetransfer, is closely related to abnormal expression and renal clear cell carcinoma andits biological behavior, the expression rate of VEGF with renal clear cell carcinoma in lower degree of differentiation, lymph node metastasis and clinical stage increasingincreased shows that it plays an important role in renal clear cell carcinoma. Theexpression of VEGF and Von Hippel–Lindau (VHL) protein were negativecorrelation. The results suggest that VHL protein is one of the important links ofindirect regulation of VEGF protein. It has important reference significance to thepathogenesis of renal clear cell carcinoma, metastasis, prognosis, through thisresearch, Von Hippel Lindau (VHL) protein may become clear cell renal carcinomatarget provide the basis for new therapeutic target.
