Objective: To investigate the feasibility to construct tissue engineering vaginalmodel with bone marrow mesenchymal stem cells(BMSCs)seeded on smallintestinal submucosa(SIS).Methods:2month-old Female Japanese rabbits weighing1.5~2.5kg were usedin the experiment.In vitro cultured rabbit BMSCs were cocultured to induce into thevaginal epithelial cells and smooth muscle cells.Both of their cells were marked by5-BrdU in vitro.The isolated cells were seeded on the same surface of SIS which cutinto1cm×1cm size.After the cells attached to the surface of the material, thecompound of the cells and the SIS was implanted into subcutaneous fascia of therabbit.The implants were removed at1,2,3and4week after operation to detect thedegree of the proliferation and differentiation of epithelial cells and smooth musclecells on the material,complexs vascular and degradation of the SIS in vivo by thehistological examination and anti-cytokeratin (CK), smooth muscle actin (SMA)immunohistochemistry and immunofluorescence check.Results: The seed cells were able to penetrate, grow into the material of SIS,And observed a variety of tissue components. At2th week after operation, the cellsproliferation layered were observed, SIS began to degrade and tissue structures weregradually clear. SIS degraded completely, and observed a large number ofangiogenesis at4th week. The labeled seed cells by5-Brdu were found in the SIS byimmunofluorescence, suggesting that implanted cells survived and participated thetissue regeneration.The smooth muscle actin (SMA) specified and cytokeratin (CK)specified immunohistochemical staining showed that the transplanted cells in vivocould differentiate.Conclusion: The tissue engineered vaginal, which is constructed withBMSC-induced cells as the seed cells SIS as the matrix,could continue to form amulti-cell structure by proliferation and differentiation.
